野菊花总黄酮对大鼠肺纤维化的干预作用

         

摘要

To study the effects and mechanism of total flavonoids of Chrysanthemum indicum ( TFC ) on pulmonary fibrosis ( PF ) in rats. Methods A total of 30 Wistar rats were randomly divided into the sham operation group, model group TFC 168 mg/kg group, TFC 336 mg/kg group and prednisone group. Rats in the model group, TFC 168 mg/kg group, TFC 336 mg/kg group group and prednisone group were injected with a single dose of Bleo-mycin A5 by trachea, respectively, and rats in sham operation group with the same normal saline. After 24 h, TFC solution of different dosages ( 168, 336 mg/kg ) was respectively given to rats in the groups daily by gastrogavage, while equal volume of normal saline was given to those in the sham operation gorup and model group, and an equal volume of prednisome ( 3. 22 mg/kg ). On the 28th days, all rats were killed, the levels of superoxide dismutase ( SOD ) and malondialdehyde ( MDA ) in the serum were determined, the determination of hydroxyproline( HYP ) in lung homogenates was analyzed, and the lung was incised to make pathological sectons which were stained with HE and Masson. The gene expression of TGF-β1 mRNA was detected by RT-PCR. Results Compared with the model group, TFC could significantly decreases the levels of SOS, MDA and concentration of HYP. TFC treated rats was also improved. Moreover TFC could decrease the expression of TGF-1 mRNA. Conclusion TFC has the inhibitory effect on PF induced by Bleomycin A5, which may be accociated with decreasing the expression of TGF-β1 mRNA.%目的 研究野菊花总黄酮(TFC)对大鼠肺纤维化(PF)的预防作用.方法 30只Wistar大鼠,随机均分为假手术对照组、模型组、TFC(168、336 mg/kg)组、强的松组.除假手术对照组外其余各组大鼠气管内一次性滴注盐酸平阳霉素诱导PF模型.次日TFC两组和强的松组分别给予TFC 168、336 mg/kg及强的松3.22 mg/kg,假手术对照组、模型组用生理盐水代替.给药后第28天处死全部大鼠.观察各组肺间质纤维化形成的病理变化;检测大鼠肺组织羟脯氨酸(HYP)、血清超氧化物歧化酶(SOD)、丙二醛(MDA)含量;RT-PCR法观察各组大鼠肺组织TGF-β1 mRNA的表达.结果 与模型组比较,TFC组病理变化减轻,TFC能显著降低大鼠肺组织HYP以及血清中SOD、MDA 的含量并能下调大鼠肺组织TGF-β1 mRNA表达.结论 TFC可以改善盐酸平阳霉素诱导的PF大鼠一般情况,并通过抑制大鼠肺组织TGF-β1 mRNA表达,以达到抗纤维化的作用.
