植物叶片中硫含量测定方法研究

         

摘要

系统研究硫酸钡比色法测定植物叶片样品中硫含量的最适宜条件。以葡萄叶片硫化物提取液为研究对象,研究保护剂的选择和用量、酸度的选择、氯化钡的形态和用量、反应温度对吸光度大小的影响。结果表明,葡萄叶片提取液2 mL在甘油-乙醇(1:1)2 mL,10 mg/mL BaCI2溶液2 mL,25% HCI溶液2 mL,常温下反应的条件下,测定其470 nm处的吸光度值,标准硫溶液在0~32μg/mL范围内与吸光度呈良好线性关系( R2﹦0.9984),平均回收率为99.17%,RSD 为1.89%。该方法简便、灵敏度高、重复性好,可用于植物叶片中硫含量的测定。%The study was to investigate an optimaI barium suIfate turbidity to determine suIfur in pIant Ieaves. The suIfate extract of grape Ieaves was used to optimize stabiIizer choice and amount,acidity rea-gent amount,form and content of BaCI2 ,reaction temperature of reaction,can affect the resuIt. ResuIt showed that a mixture of 2 mL of extract,2. 0 mL of gIycerinum-ethanoI(1 :1),2. 0 mL of 10% BaCI2 , 2. 0 mL of 25% HCI diIuted with distiIIed water at room temperature,foIIowed by detection at 470 nm. There was a good Iinearity between the absorbance and suIfur standard soIution concentration from 0 ~32 μg/mL(R2 ﹦0. 998 4). Standard recovery rate at two IeveIs was 99. 17%,with a reIative standard deviation of 1. 89%. The method was simpIe,accurate and suggested as an appropriate method for the de-termination of suIfur in pIant Ieaves.
