蛋白酶水解豆渣制备大豆肽的研究

         

摘要

以大豆豆渣为原料,分别采用木瓜蛋白酶、碱性蛋白酶、中性蛋白酶对大豆豆渣进行水解制备大豆肽.研究了3种蛋白酶在不同温度、pH值、水解时间、酶浓度下水解大豆豆渣的最适酶反应条件,以及水解产物对酸豆乳发酵的影响.结果得出,实验豆渣蛋白质含量为10.4 g/(100 g);木瓜蛋白酶的最适水解条件是底物质量分数5%,酶浓度20 000 U/g,水解温度为50℃,pH7,水解6h,最大水解度可达87.31%;碱性蛋白酶的最适水解条件是底物质量分数5%,酶浓度15 000 U/g,水解温度为55℃,pH9,水解3h,最大水解度可达91.23%;中性蛋白酶的最适水解条件是底物质量分数5%,酶浓度15 000 U/g,水解温度为45℃,pH7,水解8h后达到最大水解度,最大水解度可达81.42%.%Using bean dregs as raw material, soybean peptide was prepared using papain,alkaline protease and neutral protease, respectively. We mainly studied the optimal enzyme reaction condition of different hydrolysis temperature, pH, hydrolysis time and enzyme concentration and the effect of hydrolysate on fermented sour soybean milk. Experimental results indicated that: protein content of soybean dregs was about 10. 4 g/(100 g) ; the optimum hydrolysis conditions for papain were as that: substrate concentration of 5% (w/w) , enzyme concentration of 15 000 U/g, hydrolysis temperature of 50°C , pH7, hydrolysis time of 6 h, and maximum degree of hydrolysis (DH) was 87. 31%;The optimum hydrolysis conditions for alkaline protease were as follows: substrate concentration of 5% (w/w) , enzyme concentration of 15 000 U/g, hydrolysis temperature of 55°C , pH9, hydrolysis time of 3 h, and maximum DH was 91. 23% : The optimum hydrolysis conditions for neutral protease was as: substrate concentration of 5% (w/w) , enzyme concentration of 20 000 U/g, hydrolysis temperature of 45°C , pH7, hydrolysis time of 8 h, and maximum DH was 81. 42% .
