直链淀粉手性固定相拆分泮托拉唑钠对映体

         

摘要

建立了泮托拉唑钠对映体的高效液相色谱分析方法。在正相条件下,使用Chiralpak AD-H手性色谱柱,考察了流动相中极性调节剂的各个组分及其体积分数、流速以及柱温对泮托拉唑钠对映体拆分的影响。确定了达到最佳拆分效果的条件:流动相为正己烷∶异丙醇(体积比)=60∶40;流速0.6 mL·min-1,检测波长289 nm;柱温25℃。研究手性合成了( S)-泮托拉唑钠,对映体过量值( e. e.)达99.53%。在优化的色谱条件下,泮托拉唑钠対映体能达到基线分离且重复性好,可用于其质量控制和手性拆分的研究。%A chiral stationary phase HPLC method was established for enantiomeric separation of pantoprazole sodium. By using a chiral column Chiralpak AD-H(250mm×4. 6mm×5μm),the influences of concentration of organic solvent,flow rate and column temperature on the enantiomeric separation were investigated. The most optimal conditions was identified as follows:V( hexane)∶V (isopropanol)=60∶40;flow rate:0. 6mL·min-1;through wholly wavelength scanning to determine the detection wavelength of Pan-toprazole sodium:289nm;column temperature:25℃. The enantionary excess(e. e. )of the synthesized(S)-(-)-pantoprazole sodium was 99. 53%. under this optimized chromatographic conditions,the enantiomers of pantoprazole sodium reached a better baseline separation with good repeatability. This conditions could be available to the quality control and chiral separation of pantoprazole so-dium.
