补肾益精方对干性年龄相关性黄斑变性动物模型骨髓来源干细胞动员的影响

         

摘要

目的 评价补肾益精方对干性年龄相关性黄斑变性动物模型骨髓来源干细胞动员的影响.方法 通过尾静脉注射碘酸钠溶液的方法建立干性年龄相关性黄斑变性动物模型,比较不同组别小鼠外周血骨髓来源干细胞相对数量和相关细胞因子的改变.造模后第1d给药,中药组每日予补肾益精方水煎剂灌胃,对照组以蒸馏水灌胃,空白组不做处理.给药7d、14d、28 d后观察3组外周血骨髓来源干细胞相对数量和相关细胞因子的改变.结果 灌胃7d后,中药组外周血Lin-/Sca-1+/c-kit+细胞相对数量较对照组和空白组增高,差异有统计学意义;对照组外周血粒细胞集落刺激因子(granulocyte colony-stimulating factor,G-CSF)含量较空白组和中药组降低,差异有统计学意义;对照组和中药组外周血基质细胞衍生因子-1 α(StromalCell-Derived Factor 1 alpha,SDF-1a)及其受体趋化因子受体-4(C-X-C chemokine receptor type 4,CXCR-4)含量较空白组降低,差异有统计学意义.灌胃14d后,对照组外周血G-CSF含量仍较空白组和中药组降低,差异有统计学意义;中药组外周血SDF-1a、CXCR-4含量较对照组增加,差异有统计学意义.灌胃28 d后,中药组外周血G-CSF含量较对照组和空白组增加,差异有统计学意义;中药组外周血SDF-1a、CXCR-4含量仍较对照组增加,差异有统计学意义.结论 补肾益精方能够增加外周血BMCs、G-CSF、SDF-1a及其受体CXCR-4含量,对BMCs有一定的动员作用.其机制可能与补肾益精方增加外周血BMCs相关动员因子含量,促进BMCs从骨髓向外周血迁移有关.%OBJECTIVE To assess effect of Bushen Yijing formula on mobilizing bone marrow-derived stem cells in dry age-related macular degeneration animal model.METHODS This study used an animal model of sodium iodate (NaIO3)-induced retinal pigment epithelial (RPE) degeneration to observe variations of relative number and related cytokines of bone marrow-derived stem cells in peripheral blood.The first day after modeling,treatment group was administered with BushenYijing formula while the control group with distilled water.The normal group received no medication.Bone marrow cells (BMCs) and cytokine indicators changes in those three groups were observed on 7th day,14th day,28th day after treatment.RESULTS At the 7th day after treatment,the Lin-/Sca-1 +/c-kit+ cell level in treatment group was significantly higher than counterparts of control group and normal group in peripheral blood (PB) and the difference was of statistical significance;The granulocyte colony-stimulating factor (G-CSF) level in control group was significantly higher than treatment group and normal group in PB and the difference was statistically significant;The Stromal Cell-Derived Factor 1 alpha (SDF-1a) and chemokine receptor type 4 (CXCR-4) level in control group and treatment group was significantly lower than normal group in PB in statistical dimension.Fourteen days after treatment.The G-CSF level in the control group was still significantly higher than it in treatment group and the normal group in PB;The SDF-1a and CXCR-4 levels in the treatment group were significantly higher than the control group in PB.And these differences were all of statistical significance.Twenty-eight days after treatment,the G-CSF level intreatment group was still significantly higher than the control group and the normal group in PB;The SDF-1a and CXCR-4 levels in PB of the treatment group were still significantly higher than it of control group.And the differences between groups were all statistically significant.CONCLUSIONS Bushen Yijing formula increased BMCs,G-CSF,SDF-1a and its receptor CXCR-4 level in peripheral blood and played an important role in mobilizing BMCs to PB.The mechanism might be that Bushen Yijing formula increased BMCs mobilization related factors in PB which promoted BMCs mobilization to PB.
