Objective: To evaluate the effects of recombinant immunotoxin Hil2 - Luffin PI combined with tretinoin on proliferation, apoptosis and cell cycle of T lymphoma cell (Hut - 78 cells). Methods: Hut - 78 cells were treated with Hil2 - Luffin PI at different concentrations. Cell proliferation and apoptosis of Hut - 78 cells were assessed respectively by MTT assay and Annexin V/PI doubled - labeled flow cytometry. Results: Inhibition rates of Hut-78 cells were 21.33 ± 1.65, 28.30 ±2.42, 40.58 ± 1.69, 51.57 ±2.54, 63.15 ±2.41 and 72.45 ± 1.76, respectively, after the treatment with Hil2 - Luffin PI at concentration of 0.5, 1, 10, 20, 30 W 40 μg/Ml, combining with arotinoid ethylester of 10-7 mol/L for 48 h. Inhibition rates of Hut - 78 cells were 26. % ± 1.91, 38.05±1.64, 51.22±0.57, 64.30±2.19 and 72.48 ±2.23, respectively after the treatment with Hil2-Luffin PI of 30 (ng/Ml and arotinoid ethylester of 10-7mol/L at time of 12, 24, 36, 48 h. Apoptosis rates of Hut - 78 cells were 15.94± 1.66 and 34.89±2.11 respectively after the treatment with Hil2- Luffin PI at concentration of 1, 10 and 30 fig/Ml combining with 30μg/Ml arotinoid ethylester for 48 h. The Hut - 78 cells were treated with IL - 2Luffin PI of 30 μg/Ml plus 10" mol/L arotinoid ethylester for 48 hours, the proportion of cells in Gl phase increased and the cells in S phase decreased. Conclusion: Hil2 - Luffin PI plus arotinoid ethylester inhibits cell proliferation and induces cell apoptosis in Hut- 78 cells.%目的:评价重组免疫毒素hIL2- Lufin P1联合维A酸乙酯对皮肤T淋巴瘤细胞(Hut - 78)增殖、凋亡及细胞周期的影响.方法:将不同浓度hIL2- Luffin P1分别联合10-7 mol/L维A酸乙酯处理Hut - 78细胞,用MTr法检测细胞抑制率,流式细胞仪检测细胞凋亡及细胞周期.结果: 用浓度为0.5、1、10、20、30、40 μg/mL的hIL2- Luffin P1分别联合10-7 mol/L的芳维A酸乙酯作用Hut - 78细胞48h,对Hut- 78细胞的抑制率分别为21.33±1.65、28.30±2.42、40.58±1.69、51.57±2.54、63.15±2.41、72.45±1.76;用30μg/mL的hIL2 - Luffin P1联合10-7mol/L芳维A酸乙酯分别作用于Hut - 78细胞12、24、36、48h,对Hut - 78细胞的抑制率为26.96±1.91、38.05±1.64、51.22±0.57、64.30±2.19、72.48±2.23.经浓度为1、30μg/mL的hIL2- Luffin P1分别联合10-7 mol/L的维A酸乙酯作用Hut - 78细胞48h,细胞的凋亡率分别为15.94±1.66和34.89±2.11.经30μg/mL的hIL-2 - Luffin P1联合10-7 mol/L的维A酸乙酯作用Hut - 78细胞48h,主要表现为G1期细胞比例增加和S期减少.结论:hIL2- Luffin P1联合维A酸乙酯能够抑制Hut - 78细胞的增殖及凋亡.
