破壁灵芝孢子对小鼠移植肝癌细胞的生长和端粒酶活性的抑制作用

         

摘要

[Objective] To investigate the effects of germination activating sporoderm-broken ganoderma spores (GSGS) on the growth and telomerase activities of mouse transplanted hepatoma. [Method] Fifty NIH nude mice transplanted with hepatoma cell suspension were randomly divided into five groups: negative control, positive control groups. The experiments continued for 10 days. On the 11th day the mice were sacrificed, tumor excised, weighed and tested telomerase activity using PCR-ELISA method. [Results] The antitumor effects of GSGS at low, medium and high dose were 62.8%, 73.5% and 81% respectively. Telomerase activity (positive vs. negative) in the negative control group was 9:1; in the low dose GSGS group was 3:7 and in both the medium and high dose groups were 1:9.The differences were highly significant. [Conclusion] GSGS was shown to be a potential antitumor agent and telomerase inhibitor in tumor tissues.%目的 研究GSGS对小鼠移植肝癌细胞的生长和端粒末端转移酶作用.方法 用肝癌细胞悬液接种50只NIH裸鼠,再将其分为5组:阴性对照,阳性对照(环磷酰胺20mg/(kg·d),GSGS小剂量2g/(kg·d),中等剂量4 g/(kg·d)和大剂量8 g/(kg·d).实验持续10天,在第11天,将小鼠处死,切除肿瘤,称量并且用PCR-ELISA检测端粒末位转移酶.结果 GSGS的抗肿瘤效果在低剂量,中等剂量,大剂量分别是:62.8%、73.5%和81%.端粒末端转移酶活性(阳性比阴性)在阴性对照组是9:1;在小剂量组是3:7,在中等剂量和大剂量组都是1:9.差异有显著性.结论 证实GSGS在肿瘤组织中抗癌和端粒末端转移酶抑制剂的作用.
