[Objective]To investigate the effect of the culture medium of astrocyte on differentiation of neural stem cells (NSCs) in vitro, and explore the proper condition of neural stem cells' differentiation.[Method]The medium from cultured astrocyte of neonatal and adult SD rats were collected respectively and mixed with DMEM/F12medium according to the proportion of 1:3. Fetal brain derived NSCs were induced to differentiate in the mixed mediums. The induced cells were observed under the phase contrast microscope and were detected and counted by immunofluorescent technique.[Result]The mixed medium of neonatal astrocyte culture medium and DMEM/F12medium induced more NSCs to neurons. [Conclusion]The medium from cultured neonatal astrocyte promotes NSCs' neuronal differentiation.%目的研究新生SD大鼠星形胶质细胞培养上清液对神经干细胞(neural stem cells,NSCS)体外定向分化的影响,探讨神经干细胞分化条件.方法收集新生和成年SD大鼠星形胶质细胞培养的上清液,以1:3比例同DMEM/F12培基混合,分别对胎鼠来源的神经干细胞进行诱导分化,倒置相差显微境下观察细胞的生长情况,并采用免疫荧光检测方法对分化细胞鉴定和计数.结果新生鼠星形胶质细胞培养上清液与DMEM/F12的混合培基诱导神经干细胞分化为神经元的比例明显提高.结论新生鼠星形神经胶质细胞能促使神经干细胞向神经元方向分化.
