创面修复中骨桥蛋白和表皮生长因子受体表达变化的实验研究

         

摘要

目的:探讨烧伤后创面组织愈合过程中骨桥蛋白(osteopontin,OPN)和表皮生长因子受体(epithelial growth factor receptor,EGFr)表达顺序及分布规律.方法:应用免疫组化与原位杂交方法对临床15例烧伤患者创面愈合过程中OPN和EGFr表达变化进行检测,并计算阳性表达颗粒细胞数.结果:烧伤创面愈合中OPN和EGFr有明显规律性的表达变化,但两者的表达模式不尽相同.OPN的表达在伤后3 d达最高值,其阳性颗粒主要分布于真皮层成纤维细胞的胞浆中.EGFr的表达在伤后7 d达峰值,其阳性颗粒主要分布于表皮基底细胞胞浆及真皮成纤维细胞的胞核内.结论:烧伤可以诱导皮肤创面组织中OPN和EGFr的表达,其表达具有时相与部位分布特征.这表明OPN和EGFr作为调控因子参与了临床创面修复过程,合理调控其表达变化有助于临床创面修复.%Objective: To investigate the sequence and characteristics of expression of OPN and EGFr during the clinical wound healing. Metbods: The expression of OPN and ECFr of 15 bumed patients were examined during the wound healing by immunohistochemistry and in situ hyhridization, and the number of their positive cells was calculated. Results: In clinical wound healing, changes of OPN and EGFr expression were observed obviously , but those two proteins showed different expression models in sequence and distribution. The expression of OPN became increasing at 1 postburn day, and reached peak at 3 postbum days, the positive granules were mainly distributed in cytoplasm of fibroblasts. The expression of EGFr reached peak at 7 postbum days and the positive granules were mainly distributed in cytoplasm of basal cells and neucleus of fibroblasts. Conclusion: Thermal injury can induce OPN and EGFr expression which shows phasic control and regional distribution. Those results suggest Lhat OPN and EGFr are involved with the repair process as a regulator. It is helpful for the clinical wound healing to reasonably control OPN and EGFr expression.
