胰腺大部切除法制作大鼠高血糖模型的实验研究

         

摘要

Objective: To get hyperglycemia model of rats by subtotal pancreatectomy. Methods: Sixty rats were divided into three teams, team P+H (n=30) :resect more than 90% of pancreas, then the rats were given high-carbohydrate diet.Team P (n=20) :only receive subtotal pancreatectomy. Team H (n=10) :receive false surgery and high-carbohydrate diet.Changes of blood sugar, insulin, weight and glucose tolerance reaction were monitored, the tissue section of remnant pancreas were stained by HE and immunohistochemistry. Results: 24 hours after operation, blood sugar of most rats were rising , but there was no typical diabetic symptom observed. Morphological investigation on pancreas showed the deduction and atrophy of islet to varying degree after 2 month of operation in some of the rats which blood sugar constantly more than 7 mol/L,while the others showed hypertrophy and enhancement of insulin sLain in the islets. In the normal diet team, blood sugar gradually decreased after 1-3 d increasing, but the sugar-resist test still showed glucose intolerance. Conclusion:Stable hyperglycemia model of rats can be made by pancreatectomy and high-carbohydrate diet. Moderate hyperglycemia may be contribute to the regeneration of islet , while long-term over mcrease of blood sugar may cause over loading and injury of beta-cells.%目的:使用胰腺大部切除法制作大鼠高血糖动物模型.方法:60只SD大鼠分为三组,P+H组(n=30):切除大鼠胰腺>90%,后予高糖饮食刺激.P组(n=20):只行胰腺大部切除.H组(n=10):假手术加高糖饮食.观察术后大鼠血糖、胰岛素、体重变化及糖耐量反应并对胰腺组织切片行HE及免疫组化染色检查.结果:术后24 h起多数实验动物出现血糖升高,但未出现典型的糖尿病表现,术后2个月,部分血糖持续升高>7 mmol/L大鼠胰岛形态学检查可见有不同程度的胰岛萎缩、beta细胞数量减少,而其他大鼠则可见胰岛肥大,胰岛素染色增强.不予高糖饮食刺激组在出现一过性血糖升高后又渐回复正常,但糖耐量反应仍出现异常.结论:切除大鼠大部胰腺后,予高糖饮食刺激可制作稳定的高血糖动物模型,其中适当轻度血糖升高可能有助于促进胰岛再生,而持续较高的血糖水平则可加重胰腺损伤时胰岛负担并致其损伤.
