RP-HPLC法测定银天颗粒中主要药效成分的含量

         

摘要

目的:建立测定银天颗粒中绿原酸和栀子苷含量的方法.方法:采用反相高效液相色谱法.绿原酸检测色谱条件:色谱柱为Platisil-ODS(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸(20∶80),检测波长为326 nm,流速为1.0 mL·min -1,柱温为25℃.栀子苷检测色谱条件:色谱柱为Agilent Zorbax SB-C18(250 mm×4.6 mm,5μm),流动相为乙腈-水(10∶90),检测波长为238 nm,流速为1.0 mL·min-1,柱温为25℃.结果:绿原酸、栀子苷的检测浓度分别在5.4~108.0、11~110 μg·mL-1范国内与各自峰面积积分值呈良好线性关系(r均为0.9999);平均回收率分别为99.04% (RSD=2.66%,n=6)和97.69%( RSD=2.07%,n=6).结论:本方法专属性强、重复性好、结果准确,可用于银天颗粒的质量控制.%OBJECTIVE: To establish methods for the content determination of chlorogenic acid and gasminoidin in Yintian granules. METHODS: RP-HPLC method was adopted. The chromatographic conditions of chlorogenic acid were as follows: Plati-sil-ODS(250 mmx4.6 mm, 5 μm)column was used with mobile phase consisted of acetonitrile-0.2% phosphoric acid(20:80)at a flow rate of 1.0 mL·min‐1.The detection wavelength was set at 326 nm. The column temperature was 25 ℃. The chromatographic conditions of gasminoidin were as follows: Agilent Zorbax SB-C18(250 mm×4.6 mm, 5 um)column was used with mobile phase consisted of acetonitrile-water solution(10:90)at the flow rate of 1.0 mL·min‐1. The detection wavelength was set at 238 nm and column temperature was 25 ℃.. RESULTS: Chlorogenic acid showed a good linear relationship at range of 5.4~ 108.0 μg·mL‐1(r= 0.999 9)with an average recovery of 99.04%(RSD=2.66%,n=6). Gasminoidin showed a good linear relationship at range of 11~ 110 ug·mL‐1(r=0.999 9)with an average recovery of 97.69% (RSD=2.07% ,n=6). CONCLUSION: The methods are specific, reproducible and accurate, and can be used for the quality control of Yintian granules.
