整合素β3与肺癌A549细胞对ADM敏感性的关系

         

摘要

Objective To investigate the effect of integrin (33 on influencing the sensitivity to chemotherapy of human pulmonary adenocarcinoma A549 cells. Methods Human pulmonary adenocarcinoma A549 multicellular spheroids( MCS) were constructed with three dimensional cell culture methods. Cell counting using blood cell counter was employed to detect the sensitivity to ADM of AS49 MCS before and after blocking integrin p3. The integrin |33 expression of AS49 MCS and cell apoptosis was detected by flow cy-tometry. Results A549 MCS were successfully established. The integrin (13 expression of A549 MCS elevated with the concentration of ADM( <0.02μg/ml). Blocking integrin (33 leaded to higher sensitivity to ADM,and Icwdecreased from 0. 19μg/ml to 0. Lμg/ml, and apoptosis rate increased from( 15. 81 ± 1. 87)% to (30. 14 ± 2. 89)%. Conclusion The cell adhesion molecules integrin p3 could influence the sensitivity to chemotherapy of AS49 MCS and inhibiting cell apoptosis might be its mechanism.%目的 探讨整合素β3( Integrin β3)对人肺腺癌A549细胞系的药物敏感性的作用及可能机制.方法采用三维培养方法获得A549细胞团簇(MCS);血球计数器计数法比较阻断Integrin β3作用前后A549 MCS对阿霉素(ADM)的敏感性;流式细胞术检测Integrinβ3表达和A549细胞凋亡情况.结果经过5天细胞培养,成功建立了A549 MCS.在一定浓度范围内(<0.02 μg/ml),A549 MCS中Integrinβ3表达量随ADM浓度的增加而增加.阻断Integnin β3的作用后,A549 MCS对ADM的敏感性增强,半数抑制浓度IC50由0.19μg/ml下降至0.11 μg/ml,细胞凋亡率从(15.81±1.87)%上调至(30.14±2.89)%.结论Integrin β3的表达水平能够影响A549 MCS的药物敏感性,可能与Integrin β3抑制凋亡相关.
