缺氧诱导因子-1α在垂体腺瘤中的表达及其临床意义

摘要

目的 探讨缺氧诱导因子-1α(HIF-1α)在垂体腺瘤中的表达及与垂体腺瘤侵袭性的关系.方法 收集的42例垂体腺瘤标本中侵袭性垂体腺瘤27例,非侵袭性垂体腺瘤15例,应用逆转录聚合酶链反应(RT-PCR)检测HIF-1αmRNA的表达;免疫组化染色检测HIF-1α蛋白的表达.并对侵袭组与非侵袭组的HIF-1α蛋白、HIF-1αmRNA表达及不同大小肿瘤HIF-1α蛋白表达进行分析比较.结果 非侵袭组垂体腺瘤中HIF-1α蛋白表达阳性率与侵袭组比较,差异有统计学意义(χ2=9.15,P=0.005);而HIF-1αmRNA在侵袭组的表达水平与非侵袭组比较,差异无统计学意义(t=1.041,P=0.329);垂体腺瘤直径>30mm者HIF-1α蛋白表达阳性率与直径10~30 mm者比较,差异有统计学意义(χ2=4.89,P=0.041).结论 HIF-1α蛋白在侵袭性垂体腺瘤中的表达增加,并且与垂体腺瘤侵袭行为的产生有关.%To explore mRNA and protein expressions of hypoxia - inducible factor - 1 a ( HIF - 1 a ) in pituitary adenomas and its relationship with invasion of pituitary adenomas. Methods Among the collected 42 cases of pituitary ade-rntuitary adenomas and its relationship with invasion of potuitary adenomas Methods Among the collected 45 cases of pituitary ade- rnnomas, 27 cases were invasive pituitary adenomas and 15 cases were non — in v asrv e pituiTary adenomas. Reverse Transcription poi—rnymerase chain reaction ( RT - PCH ) was used to determine the mRNA expression of HIF - 1 a. Immunohistochemistry staining was used to determine the protein expression of HIF - 1 α. The mRNA and protein expressions of invasive pituitary adenomas group and non — invasive pituitary adenomas group and the protein expression of HIF — 1 ex in pituitary adenomas with different size were ana-lyzed and compared. Results The positive rate of protein expression of HIF - 1α in invasive pituitary adenomas group showed statistically significant difference compared with that of the non - invasive pituitary adenomas group ( x2 =9. 15, P = 0. 005 ). While the mRNA expression of HIF - 1 α in invasive pituitary adenomas group showed no statistically significant difference compared with non - invasive pituitary adenomas group ( t = 1. 041 , P = 0. 329 ) . The positive rate of protein expression of HIF - 1α in pituitary adenomas with diameter larger than 30 mm showed statistically significant difference compared with those with diameters between 10 mm to 30 mm ( X2 =4.89, P = 0.041 ) . Conclusion There is overexpression of HIF- la protein in pituitary adenomas, and these studies suggest that the activation of HIF - 1 a protein can increase the invasion of pituitary adenomas.
