Objective To investigate the expression of Shh and Ptch in prostate cancer, analyze its impact on pathological features and explore its effect on occurrence and progression of prostate cancer. Methods 100 cases of prostate cancer were studied by immunohistochemisty using the Shh and Ptch antibody. Results (1)The expression of Shh and Ptch in prostate cancer was markedly elevated compared with their corresponding adjacent normal tissues, and the expression of Shh and Ptch has positive correlation; (2)The expression of Shh and Ptch was associated with pathological grades(Gleason score)(P<0.05); (3)The expression of Shh and Ptch in prostate cancer has nothing to do with preoperative PSA(P>0.05). Conclusion Higher expression of Shh and Ptch activates the Hedgehog signaling pathway in prostate cancer, and the expression of Shh and Ptch is related to the pathological grade, intimately relevant to occurrence and progression of prostate cancer, Shh, Ptch may be the ideal targets in diagnosis and treatment of prostate cancer.%目的 检测hedgehog (Hh)信号通路中的Shh和Ptch蛋白在前列腺癌(PCa)中的表达,分析其与病理特征的关系,探讨两者在PCa发生、发展中的作用.方法 采用免疫组织化学染色方法检测100例PCa标本及癌旁正常组织中Shh及Ptch的表达.结果(1) Shh和Ptch在PCa中较癌旁正常组织呈高表达,且两者呈正相关;(2) PCa组织中Shh、Ptch的表达程度与病理分级(Gleason分级)之间有显著相关性(P<0.05); (3) PCa组织中Shh与Ptch表达程度与患者术前PSA无明显相关性(P>0.05).结论PCa中Shh和Ptch的高表达激活了Hh信号通路,且Shh和Ptch表达程度与病理分级相关,与PCa的发生发展密切相关,且为协同作用,Shh、Ptch有可能成为PCa诊断及治疗的理想分子靶点.
