Objective To investigate the effects of testosterone on the released non-exocytosis norepinephrine of the isolated rat heart. Methods Sprague-Dtawley male rats (n=40) were randomly divided into the testosterone group and the control group. The control group was primed with modified K-H buffer, and the testosterone group was primed with modified K-H buffer containing different concentrations of testosterone (100.0 nmol/L, 10.0 nmol/L and 1.0nmol/L). The level of the overflowed norepinephrine was measured by high pressure liquid chromatography and electrochemical detection (HPLC-EC). Results Under acute ischemia, the release of norepinephrine in the testosterone group (1.0 nmol/L, 10.0 nmol/L and 100.0 nmol/L) waa significantly lower than that in the control group (P<0.0l) Conclusion Testosterone might inhibit the release of norepinepherine of the isolated rat heart.%目的 研究睾酮对离体大鼠心脏去甲肾上腺素非出胞释放的影响.方法 正常雄性SD大鼠40只,随机分为空白对照组和实验组,观察缺血时去甲肾上腺素的释放.空白组用不含任何药物的K-H液灌注；实验组为睾酮高、中、低浓度组,分别用含100.0nmol/L、10.0nmol/L、1.0 nmol/L睾酮的K-H液灌注.用高效液相色谱-电化学法(HPLC-EC)测定心脏流出液中去甲肾上腺素的浓度.结果 心脏缺血时,空白组和睾酮组比较,睾酮1.0 nmol/L,10.0 nmol/L和100.0 nmol/L组可减少心脏NE释放(P＜0.01).结论 睾酮可抑制离体心脏去甲肾上腺素的非出胞释放.
