“肝肾精血亏虚”大鼠动物模型的建立

         

摘要

目的:研究“肝肾同源于精血”的病理生理机制、肝肾精血亏虚证本质及药物筛选,建立“肝肾精血亏虚”大鼠动物模型。方法:于Wistar大鼠出生后第2、4、6、8、10d皮下注射左旋谷氨酸单钠(MSG)每次4mg/g体重,第8周手术切除MSG-大鼠大部分(约68%)肝脏。结果:“肝肾精血亏虚大鼠”的肝再生度、肝细胞分裂指数(%)、肝重/体重比值、骨髓有核细胞增生度、外周血红细胞计数和血红蛋白均显著低于正常和生理盐水对照组(P<0.01)。经补益肝肾精血的左归丸治疗后,“肝肾精血亏虚大鼠”上述指标显著改善(P<0.05,或P<0.01)。结论:大部切除MSG-大鼠肝脏可制作“肝肾精血亏虚大鼠”动物模型,肝再生能力可作为观察肝肾精血亏虚证的客观化指标。%Objective In order to study the mechanism of “The liver and the kidney have a common source of the essence and the blood”,establish the model of rat of deficiency of the kidney-essence and the liver-blood. Method Litters delivered Wistar rats were treated with monosodium glutamate(MSG) dissolved in saline solution. MSG 4mg/g b.wt. was injected s.c. daily from the 2nd to the 10th day after birth. On the 8 weeks, partial hepatectomy was performed by excision of the median and left hepatic lobes according to the method of Higgins and Anderson under pentobarbital anesthesia. Results The degree of liver regeration, MI, the rate of liver weight/body weight, the degree of proliferation of nucleated cells in bone marrow, RBC and Hb were significantly lower than the normal and saline solution control group. Conclusion MSG-rat partial hepatectomy was performed by excision of the median and left hepatic lobes is the model of rat of deficiency of the kidney-essence and the liver-blood.
