血红素氧合酶-1对大鼠肝脏缺血再灌注NF-κB p65和细胞凋亡的影响

         

摘要

Objective To explore the effects of Heme Oxygenase-1 on NF-κB P65 and apoptosis in rats dur-ing the liver ischemia/eperfusion damage.Methods SD rats were randomly divided into five groups:normal control group,sham group,I/R group, I/R+hemin group,and I/R+ZnPP group.Blood samples were collected from the postcava at 6h and12h after the operation,and the serum was isolated to measure alanine aminotransferase (ALT) and aspartate aminotransferase ( AST) .The expressions of NF-κBp65 and Fas protein in livers of rats were examined by westernblotandimmunohistochemistry.Results ①ThelevelsofALTandASTweresignificantlylowerinhemin group than those in I/R group and ZnPP group( P<0.01) .②The expression of NF-κB p65 was lower in hemin group than those in I/R group(P <0.01);the expression of NF-κB p65 was higher in ZnPP group than those in I/R group (P<0.05).③The expression of Fas protein was markedly increased in ZnPP group than those in hemin group(P<0.01).Conclusion Protective effects of HO-1 against hepatic ischemia/reperfusion damage were closely related to the reduced NF-κB P65 expression,which maybe decreased the apoptosis via inhibit activity of NF-κBp65.%目的探讨血红素氧合酶-1(HO-1)对大鼠肝脏缺血再灌注NF-κB P65、细胞凋亡的影响。方法SD大鼠采用随机数字表法分成5组:正常组(N),假手术组(S),手术组(I/R建立肝脏缺血再灌注损伤模型),手术给药Ⅰ组( I/R+hemin),手术给药Ⅱ组( I/R+ZnPP),手术后6 h、12 h下腔静脉取血分离血清检测血清谷草转氨酶( AST)、谷丙转氨酶( ALT)。 Western blot方法测定肝脏NF-κB p65的表达,免疫组化方法测定肝脏Fas蛋白的表达。结果①hemin组ALT、AST较I/R组及ZnPP组明显降低(P<0.01);②肝组织NF-κBp65的表达:hemin组NF-κB p65表达比同期I/R组减少(P<0.01);ZnPP组NF-κB p65表达比同期I/R组增多(P<0.01);③肝组织Fas:hemin组Fas表达减少(P<0.05),ZnPP组Fas表达明显增多(P<0.01)。结论 HO-1对肝脏缺血/再灌注损伤的保护作用与NF-κBP65密切相关,可能通过抑制NF-κB p65的活性来减少细胞凋亡。
