实验性2型糖尿病恒河猴模型的构建及其部分临床特征分析

摘要

目的 构建2型糖尿病(T2DM)恒河猴模型,使之成为研究人类T2DM的有效替身.方法 以高糖高脂饮食为基础,在出现高脂血症和肥胖状态后注射35 mg/kg的链脲佐菌素(STZ),测定体重指数、血脂、空腹血糖、胰岛素、胰岛素抵抗指数、尿糖和口服葡萄糖耐量试验等,分析其部分临床特征.结果 T2DM模型组体重指数(BMI)大于35达到重度肥胖,有高脂血症的特点,空腹血糖、胰岛素和胰岛素抵抗指数显著增高(P＜0.01),尿糖检测呈阳性,葡萄糖耐量受损并且空腹血糖高于7 mmol/L、2h的血糖水平高于11.1 mmoL/L,胰岛有轻度损伤和病变.结论 通过部分临床特征分析,T2DM模型组具有典型的T2DM临床特征,可成为T2DM研究的有效模型.%Objective To establish a type 2 diabetic (T2DM) model in rhesus monkey,and provide an effective substitute for the study of human T2DM.Methods Twelve adult rhesus monkeys aged 6-10 years were divided into three groups:T2DM group,high-fat diet group and normal control group,4 monkeys in each group.Based on high-glucose and fat diet feeding,to give streptozotocin (STZ) injection in a dose of 35 mg/kg body weight after the appearance of hyperlipidemia and obesity status in the monkeys.The body mass index (BMI),lipid profile,fasting blood glucose,insulin,insulin resistance index,urine sugar and oral glucose tolerance were measured,and some clinical features in the monkeys were analyzed.Results The BMI higher than 35 was considered as severe obesity,showing characteristics of hyperlipidemia,with significantly elevated blood glucose,insulin and insulin resistance index (P ＜ 0.01),positive urine sugar,impaired glucose tolerance,and having fasting glucose higher than 7 mmol/L and two-hour glucose level higher than 11.1 mmol/L,and had mild pathological alterations of pancreatic islets in the T2DM model group.Conclusions Analysis of some clinical features shows that the monkey models exhibit typical clinical features of T2DM,therefore,they may become effective models in human T2DM research.
