目的:探讨尼古丁对体外培养人牙龈成纤维细胞(HGFs)的增殖及其分泌IL-8的影响。方法:组织块法原代培养HGFs,MTT法观察10、50、100、250、500、1000、2000μg/mL不同浓度尼古丁对其增殖的影响;取第4代HGFs分别与10、50、100、250、500、1000μg/mL不同浓度尼古丁共同培养72 h,用酶联免疫吸附(ELISA)法检测HGFs培养上清中IL-8的含量。结果:50~2000μg/mL不同浓度的尼古丁均能明显抑制HGFs的增殖(P<0.05),且呈浓度依赖性;10~500μg/mL尼古丁均能明显促进HGFs合成IL-8(P<0.05),其中以100μg/mL浓度组的促进作用最强。结论:一定浓度尼古丁可抑制HGFs的增殖并促进其合成IL-8,提示尼古丁可能通过调控IL-8的表达而造成对牙周组织的破坏。%AIM:To investigate the effects of nicotine on the proliferation and IL-8 expression of human gingival fibroblasts(HGFs).METHODS:HGFs were primarily cultured and identified.MTT method was used to ex-amine the viability of the cells stimulated by nicotine at the concentrations of 10-2000 μg/mL respectively for 72 h. ELISA was used to detect IL-8 concentration in the culture supernatant after 72 h stimulation .RESULTS:Nicotine dose-dependently inhibited the proliferation of HGFs (P<0.05).10-500μg/mL nicotine stimulated IL-8 secretion in HGFs,100μg/mL nicotine showed the most significant stimulating effect (P<0.05 ).CONCLUSION:Nicotine can inhibit the proliferation of HGFs and increase IL-8 production by HGFs,indicating that nicotine may play a role in the progression of periodontitis in tobacco users.
