针刺治疗对躁狂模型大鼠行为学及睡眠潜伏期的影响

         

摘要

Objective To observe the effect of acupuncture treatment on behavior of manic in rat model and sleep la-tency, and explore the antimanic effects and mechanism of acupuncture therapy.Methods 100 SD rats according to the ran-dom number table method were randomly divided into blank control group, model group, sodium valproate group, acupuncture group and acupuncture not at acupoint group, 20 rats in each group.The modified multiple platform method of sleep depriva-tion ( MMPM) was used to establish the model of mania.The corresponding intervention duration was 7 d in each group.At the end of the experiment, observed rats'mental and behavior, the record the time from out of the water environment to first go to sleep (sleep latency) time, and performed the open field test.Results Compared with the blank control group, model group, sodium valproate group and acupuncture not at acupoint group’s activity level score were significantly decreased, sleep latency was significantly prolonged, the differences were statistically significant ( P <0.01 ) .Compared with the model group, sodium valproate group and acupuncture not at acupoint group’ s activity score’ s difference were not statistical signifi-cance ( P >0.05), and the acupuncture group activity level score significantly increased ( P <0.05).Compared with the model group, three treatment groups’ sleep latency were significantly shortened, differences were statistically significant ( P <0.01);three treatment group’s pairwise comparison showed that acupuncture group and sodium valproate group’s sleep laten-cy were significantly shortened than acupuncture not at acupoint group ( P <0.01 or P <0.05) , and between the acupunc-ture group and sodium valproate group showed no significant differences ( P >0.05).Conclusion Acupuncture can effec-tively improve the sleep deprivation manic model’ s prolonged sleep latency, and the effect was similar as sodium valproate, and to a certain extent can reduce the partial behavioral inhibition that due to the long-term sleep deprivation caused by the physical consumption, and its effect is better than that of sodium valproate and non acupoint stimulation.%目的:研究针刺治疗对躁狂模型大鼠行为学及睡眠潜伏期的影响及其作用机制。方法100只SD大鼠按照随机数字表法随机分为空白对照组、模型组、丙戊酸钠组、针刺穴位组和针刺非穴组,每组20只。采用改良多平台睡眠剥夺法( MMPM)建立躁狂模型。各组进行相应干预7 d。实验结束后观察各组大鼠的精神、行为,记录脱离水环境到第一次进入睡眠的时间(即睡眠潜伏期),并进行旷场实验。结果与空白对照组比较,模型组、丙戊酸钠组和针刺非穴组的水平活动得分均显著降低,睡眠潜伏期显著延长,差异均有统计学意义( P <0北.01)。与模型组比较,丙戊酸钠组和针刺非穴组的水平活动得分差异无统计学意义( P >0.05),而针刺穴位组的水平活动得分显著增高( P <0.05)。与模型组比较,3个治疗组的睡眠潜伏期均显著缩短,差异均有统计学意义( P <0.01);3个治疗组间两两比较,针刺穴位组与丙戊酸钠组睡眠潜伏期均较针刺非穴组显著缩短( P <0.01或P <0.05),而针刺穴位组与丙戊酸钠组间差异无统计学意义( P >0.05)。结论针刺穴位能有效改善睡眠剥夺躁狂大鼠模型的睡眠潜伏期,其效果与丙戊酸钠相当,并能减轻部分行为抑制,其效果优于丙戊酸钠。
