光甘草定对人皮肤成纤维细胞的作用研究

摘要

Objective This study was designed to elucidate the effects of phytoestrogen Glabridin on the proliferation of human skin fibroblast (HSF).Method HSFs were divided into blank control group (only single cell suspension was added), positive control group (treated by 10μmol/L 17β-estradiol) and experience group (0.1, 1, 10 and 100μmol/L Glabridin). After 72 hours, the Cell Counting Kit-8 (CCK-8) assay and 5-ethynyl-2'-deoxyuridine (EdU) incorporation assay were performed to assess cell viability, and the phase of cell generation cycle was determined byflow cytometry.Result The optical density (OD) of 10μmol/L experimental group and positive control group were significantly higher than that of blank control group (P<0.05); OD of 100μmol/L experimental group was significantly lower than that of blank control group (P<0.05), butthere was no significant difference in OD between 10μmol/L experimental group and positive control group (P>0.05). The percentage of EdU labeled positive cells in 10μmol/L experimental group and positive control group were significantly higher than that in blank control group (P<0.05), but there was no significant difference in percentage of EdU labeled positive cells between 10μmol/L experimental group and positive control group (P>0.05). The proportion of S phase in 10μmol/L experimental group and positive control group were significantly higher than that of blank control group (P<0.05), but there was no significant difference in proportion of S phase between 10μmol/L experimental group and positive control group (P>0.05). Conclusion Glabridin promote the proliferation of HSF.%目的 探讨光甘草定对人皮肤成纤维细胞的作用.方法 将体外培养的人皮肤成纤维细胞分为空白对照组(仅加入单细胞悬液)、阳性对照组(10 nmol/L的17β-雌二醇处理)及实验组(光甘草定浓度分别为:0.1、1、10、100μmol/L),作用72小时后,采用活细胞计数试剂盒-8(CCK-8)检测法、5-乙炔基-2'-脱氧尿苷(5-ethynyl-2'-deoxyuridine,EdU)掺入法检测成纤维细胞增殖活性;流式细胞术检测细胞S期所占比例.结果 10μmol/L实验组和阳性对照组的光密度均明显高于空白对照组(P<0.05),100μmol/L实验组光密度明显低于空白对照组(P<0.05),但10μmol/L实验组和阳性对照组光密度比较无显著差异(P>0.05).10μmol/L实验组和阳性对照组的EdU标记的阳性细胞百分比明显高于空白对照组(P<0.05),但10μmol/L实验组和阳性对照组EdU标记的阳性细胞百分比无显著差异(P>0.05).10μmol/L实验组和阳性对照组的S期占比明显高于空白对照组(P<0.05),但10μmol/L实验组和阳性对照组S期占比无显著差异(P>0.05).结论 光甘草定可以促进人皮肤成纤维细胞的增殖.
