Objective To study the relationship between progressive AAA and inflammation in rabbits according to their 18F-FDG PET/CT imaging.Methods An infrarenal AAA model of rabbits was established by intracavity elastase perfusion.Ten rabbits were killed at weeks 2 and 4 respectively after ultrasonography and PET/CT scanning.Ear venous blood samples were taken for detecting the expression of MMP-2 and MMP-9,artery tissue samples were taken for calculating the average standard uptake value,SUV and standard TBR with pathological and immunohistochemical staining.Results The maximal growth rate of aneurysm diameter was 89.21%±0.02% higher at week 2 after ultrasonography and PET/CT scanning than before ultrasonography and PET/CT scanning and was 5.80%±0.01% higher at week 4 after ultrasonography and PET/CT scanning than at week 2 after ultrasonography and PET/CT scanning.The expression levels of MMP-2 and MMP-9 were significantly higher at week 4 after ultrasonography and PET/CT scanning than at week 2 after ultrasonography and PET/CT scanning (P<0.05) and were significantly lower at week 4 after ultrasonography and PET/CT scanning than at week 2 after ultrasonography and PET/CT scanning (P<0.05).The SUVmax was significantly lower at week 4 after ultrasonography and PET/CT scanning than at week 2 after ultrasonography and PET/CT scanning (P<0.05).The rate of stained macrophage area was significantly higher at week 2 after ultrasonography and PET/CT scanning than at week 4 after ultrasonography and PET/CT scanning (P<0.05).Conclusion Inflammation plays an important role in early progressive AAA.%目的 利用18F-氟脱氧葡萄糖(18F-FDG)正电子发射断层显像(PET)/CT扫描,探讨兔腹主动脉瘤进展与炎症的关系.方法 20只新西兰雄白兔给予弹力蛋白酶腔内灌注,制备肾下腹主动脉瘤模型,术后2周和4周各取10只白兔行超声、PET/CT扫描,耳缘静脉采血后处死,检测基质金属蛋白酶(MMP)-2及MMP-9,留取动脉组织标本进行病理及免疫组织化学染色,计算最大标准化摄取值(SUVmax)及标准化靶-本底比.结果 与术前比较,术后2周动脉瘤最大直径增长率为(89.21±0.02)%;与术后2周比较,术后4周动脉瘤最大直径增长率为(15.80±0.01)%.与术前比较,术后2周MMP-2、MMP-9明显升高(P<0.05);与术后2周比较,术后4周MMP-2、MMP-9明显降低(P<0.05).术后4周SUVmax较术后2周明显降低(0.35±0.05 vs 0.90±0.03,P<0.05).术后2周巨噬细胞染色面积百分比明显高于术后4周(P<0.05).结论 炎症在腹主动脉瘤早期进展中起重要作用.
