山茱萸果核水提取物对D-半乳糖致衰老模型小鼠抗氧化能力的影响

         

摘要

Objective To discuss the effect of water extract of fructus corni officinalis core on the antioxidant capacity of aged mice model induced by D-galactose.Methods 50 male Kunming mice were randomly divided into normal control ,model and high(12 g/kg),mid-dle(6 g/kg),low(3 g/kg) dose extract groups.Morris water maze test was used to test the abilities of learning and memory .The xanthine oxi-dase method was used to measure the activity of superoxide enzyme (SOD),glucosinolates barbituric acid method was used to determine the activity of malondialdehyde (MDA),colorimetric method was used to determine the activity of total antioxidant capacity (T-AOC),DTNB method was used to determine the activity of glutathione peroxidase ( GSH-Px) .Results The average escape latencies in extract groups were significantly lower than that of model group ,the times through the platform were higher than that of model group (P<0.05).The MDA levels in extract groups were significantly lower than that of model group ,the levels of T-AOC,GSH-PX activity were higher(P<0.05).The SOD ac-tivity of extract groups were higher than that in model group ,but there were no significant differences (P>0.05).Conclusions The water ex-tract of fructus corni officinalis core could improve the antioxidant abilities of brain tissue and improve the abilities of learning and memory in aging mice.%目的:探讨山茱萸果核水提取物对D-半乳糖致衰老模型小鼠抗氧化能力的影响。方法选择50只昆明雄性小鼠随机分为正常对照组(10只)、模型组(10只)和提取物组(30只)。其中提取物组依据提取物剂量分为高剂量组(12 g· kg-1· d-1)、中剂量组(6 g· kg-1· d-1)、低剂量组(3 g· kg-1· d-1),每组10只。正常对照组腹腔注射生理盐水,其余各组通过皮下连续8 w 注射500 mg· kg-1· d-1D-半乳糖制备衰老模型。采用Morris水迷宫检测各组小鼠的学习记忆能力,完成测试后采用黄嘌呤氧化酶法测超氧化物歧化酶( SOD)活性,硫代巴比妥酸法测定丙二醛(MDA),比色法测定总抗氧化能力(T-AOC),DTNB法测定谷胱甘肽过氧化物酶(GSH-Px)。统计分析所有小鼠5 d 内 Morris水迷宫实验逃避潜伏期、穿越平台次数和脑组织MDA、SOD、GSH-Px、T-AOC含量。结果提取物组小鼠平均逃避潜伏期明显低于模型组,前者穿越平台次数高于后者(P<0.05);但各剂量组间平均逃避潜伏期,穿越平台次数无显著差异(P>0.05)。提取物组小鼠MDA水平明显低于模型组,前者T-AOC、GSH-Px活性高于后两者(P<0.05);提取物组小鼠SOD活性高于模型组,但无显著差异性(P>0.05)。结论山茱萸果核水提取物可能通过清除 D-半乳糖模型小鼠产生过多的自由基,进而提高脑组织的抗氧化能力,从而起到延缓衰老的作用及改善衰老小鼠的学习记忆能力,中等剂量的效果最好。
