决明子对家蚕寿命的影响及机制

         

摘要

Objective To explore the antioxidant activity and anti-aging effect of Semen cassiae by observing Bombyx mori L ’ s life span driven by free-radical theory.Methods The antioxidant capacity of Semen cassiae extracting by distilled H 2O and 100%ethanol were evaluated with 1,1-Diphenyl-2-picrylhydrazyl radical-2,2-Diphenyl.1-(2,4,6-trinitrophenyl) hydrazyl (DPPH・ ) spectrometry compared with vitamin E (VE) using 96 micro plate,IC50 as a standard.If IC50 was lower,that meant the antioxidant capacity was higher .Results In the experiment of in vitro,the antioxidant capacity of vitamin E was highest ,the Semen cassiae water extract was higher ,Semen cassiae 100%ethanol extract was lowest .So the Semen cassiae water extract was selected to use in vivo .During experiment ,the Bombyx mori L.with the same age and growth were divided into five groups ,included that control group,high dose (0.65 g/ml),middle dose (0.35 g/ml),low dose (0.15 g/ml) and positive group (VE),the sample was 85 of each group.Physic liquor was daubed on the mulberry leaf ,and was given to the Bombyx mori L.3 times during 4 instars and 6 times during 5 instars.At third day of 5 instars,the Haemolymphe was collected ,and SOD activity,GSH-Px activity,MDA concentration were measured by ELISA as antioxidant data .The results showed that the life-span of Bom-byx mori L’ s larva was significantly longer in high ,middle,low dose group compared with control group .The extend percentage of whole life span for each group were 15.22%in high dosage group ,12.31%in middle dosage group ,9.06%in low dosage group respectively .There was dose-effect relationship in the experimental group .Different concentration drug had significant effect on GSH-Px, SOD, MDA.GSH-Px and SOD were significantly increased ,MDA was significantly decreased in high and middle dose groups .There was significantly relationship be-tween the SOD activity and the life span of Bombyx mori L .(r=0.971,P<0.01).Conclusions The life span of Bombyx mori L.could be extended by Semen cassiae water extract through the enhancement of SOD activity which is consistent with prediction and supported the free -radical theory.%目的:探讨决明子对家蚕寿命的影响及机制。方法不同溶媒超声提取决明子制备溶液,96孔板测定抗氧化活性,另取龄期相同、生长整齐的三眠蚕随机分为五组,即空白对照组,决明子高(0.65 g/ml)、中(0.35 g/ml)、低(0.15 g/ml)剂量组,维生素 E组,每组85条。使用涂抹法将不同浓度药液涂抹于新鲜桑叶上,4龄期内喂食3次,5龄期内喂食6次。期间称体重,观察动物生长情况。在5龄期第3天采集血淋巴,测超氧化物歧化酶( SOD)、谷胱甘肽过氧化物酶( GSH-Px)及丙二醛( MDA)等抗氧化指标的活性。结果维生素E 的体外抗氧化活性>决明子水溶液>决明子醇溶液;家蚕实验中,与空白对照组相比,决明子高、中、低剂量处理显著延长家蚕各生长阶段天数( P<0.05),对全生长期的延长率分别为15.22%、12.31%、9.06%,均达到了极显著的差异水平,且具有一定的量效关系。不同生药浓度决明子处理对家蚕血淋巴中 GSH-Px、SOD、MDA的影响均有极显著差异(P<0.001),且GSH-Px、SOD在中剂量组和高剂量组有显著增高,MDA有显著降低;血清中SOD活力与家蚕全生长期间有极显著正相关关系( r=0.971,P=0.006)。结论决明子有助于家蚕体内抗氧化能力的提高。
