眼针对大鼠脑缺血再灌注损伤不同时间点海马组织TNF-α表达的影响

         

摘要

Objective To investigate TNF-a expression at different time points in the brain of rats with cerebral ischemia-reperfurnsion(IR) injury, in order to discuss the mechanism of eye-acupuncture therapy and observe the intervention effects on the hippocampus. Methods Healthy SD rats, male and female, were randomly divided into normal, sham-operated control, model and eye-acupuncture groups. Sham-operated control, model and eye-acupuncture groups were divided into 3 , 24 and 72 h subgroups. The rat model of cerebral IR was established by suture method. Eye acupoint liver area, kidney area, upper jiao area and lower jiao area were selected and acupunctured horizontally, inserting needle in 3 mm deep, retaining needle for 20 min. All the animals were executed respectively at 3, 24, 72 h after reperfusion and draw materials. The method of immunohistochemistry and Western blot technology were taken to detect the changes of hippocampus TNF-a expression in rats after the eye-acupuncture therapy. Results Compared with that of model group, TNF-a expression in the hippocampus of rats of the eye-acupuncture group was significantly decreased obviously at 3, 24, 72 h after reperfusion. Conclusions The eye-acupuncture therapy could down-regulate hippocampus TNF-a expression in rats with cerebral IR injury, suppress inflammatory response induced by cerebral IR and play a role in protecting brain.%目的 观察眼针对大鼠脑缺血再灌注后肿瘤坏死因子-α( TNF-α)蛋白表达的影响,以探讨眼针对缺血再灌注大鼠脑海马区细胞的保护作用及其作用机制.方法 健康SD大鼠,雌雄不拘,设正常对照组、假手术组、模型组、眼针组;采用改良的线栓法复制大鼠大脑中动脉缺血再灌注损伤模型,选取眼穴肝区、肾区、上焦区、下焦区,平刺,进针3 mm,留针20 min;分别于脑缺血再灌注后3、24、72 h处死动物并取材;应用蛋白质印迹法(Western印迹)检测海马组织中TNF-α蛋白的表达.结果 眼针组海马组织TNF-α在脑缺血再灌注后3、24、72 h均较模型组明显降低,差异显著.结论 眼针疗法可以降低脑缺血再灌注模型大鼠海马组织TNF-α的表达,进而抑制由脑缺血再灌注诱导的炎症反应,发挥脑保护作用.
