氟伐他汀对心肌梗死后大鼠心肌细胞Fas蛋白表达及心室重塑的影响

         

摘要

Objective To study the effects of fluvastatin on expression of Fas mRNA and ventricular remodeling in myocardial infarction rats. Methods 30 SD rats were randomized into sham-operated, saline and fluvastatin groups. Parameters of ventricular remodeling and hemodynamics were measured and the collagen component was detected in Masson stained myocardial infarction sections, as well as Fas/FasL protein and apoptosis cell ratio in myocardial tissue were measured by immunohistochemical staining method. Results Compared with the sham-operated group and saline group, the number of collagen fibers were decreased in fluvastatin group ( P < 0. 05 ). Compared with sham-operated group and saline group Fas protein expression in fluvastatin group was reduced ( P < 0. 05 ). There were obvious difference in apoptosis index of myocardial cell among sham-operated, saline and fluvastatin groups ( P < 0. 05 ). Conclusions Fluvastatin can effectively prevent post-myocardial infarction acute myocardial apoptosis and the formation of collagen fibers through Fas/FasL system and suppress ventricular remodeling.%目的 研究氟伐他汀预处理对心肌梗死后鼠心肌细胞Fas基因表达及心室重塑的影响.方法 将30只SD大鼠梗死模型随机分成假手术组、生理盐水组及氟伐他汀组.8 w后测定大鼠的血流动力学变化,并采用Masson染色法分别检测心肌梗死后胶原成分来评估心室重塑情况,同时利用免疫组化法测定Fas蛋白阳性染色指数以衡量细胞凋亡情况.结果 8 w后与假手术组比较,生理盐水组梗死区及梗死周边区胶原纤维数量明显增加,而氟伐他汀组相应部分的胶原纤维数量减少,差异显著(P<0.05).氟伐他汀组Fas蛋白阳性表达的细胞明显减少,与生理盐水组和假手术组差异显著(P<0.05).氟伐他汀组心肌细胞凋亡指数与生理盐水组差异显著(P<0.05).结论 氟伐他汀能通过Fas/FasL系统抑制梗死后鼠心肌细胞的凋亡并阻止胶原纤维的形成,从而抑制心室重塑.
