眼针对急性脑缺血再灌注损伤模型大鼠脑组织脑源性神经因子表达的影响

         

摘要

Objective To investigate the effect of eye acupuncture therapy on cerebral brain-derived neurotrophic factor (BDNF) expression in rats with cerebral ischemia-reperfusion injury and the related mechanism. Methods Rat model of cerebral ischemia-reperfu-sion was established by suture method. The methods of immunohistochemistry staining, Western-blotting and RQ-PCR were taken to detect the alteration of the BDNF protein and BDNF mRNA in rat brains after eye acupuncture therapy. Results Compared with the model group, the neurologic impairment score of the eye-acupuncture group was decreased obviously and the expression of the BDNF protein and BDNF mRNA in rat brains after eye acupuncture therapy was strongly increased( P < 0. 05 ). Conclusions Eye acupuncture therapy can reduce the cerebral ischemia-reperfusion injury evidently by increasing the expression of the cerebral BDNF protein.%目的 探讨眼针治疗急性脑缺血再灌注损伤的机制.方法 应用线栓法复制急性脑缺血再灌注损伤大鼠模型,采用眼针治疗,于再灌注后3h进行神经功能缺损评分,采用免疫组化、Western印迹、实时荧光定量聚合酶链反应(RQ-PCR)方法测定脑组织脑源性神经因子(BDNF)蛋白及mRNA表达的变化.结果 与模型组比较,眼针组大鼠神经功能缺损评分明显下降,脑组织BDNF蛋白及mRNA表达明显上调(P<0.05).结论 眼针具有明显改善大鼠脑缺血再灌注损伤的作用;其机制与上调脑组织BDNF表达有关.
