通脉地仙丸对大鼠缺血心肌血管生成及其相关基因表达的影响

         

摘要

Objective To observe the influence of Tongmai Dixian pills on the ischemic myocardial angiogenesis and its related gene expression. Methods Rats of chronic myocardial ischemia model, except the sham operation group, were randomized into model group, Shexiang Baoxin pills group and Tongmai Dixian pills high-, medium and low-dose group, and intragastric administration for six weeks. The content of VEGF and bFGF protein in the ischemic myocardial tissue, and VEGF and bFGF mRNA expression were detected by immunohistochemical staining. Results Compared with sham operation group, VEGF and bFGF protein expression of model group, Tongmai Dixian pills group and Shexiang Baoxin pills group increased (P<0.05, P<0.01, P<0.001). Compared with model group, VEGF and bFGF of Tongmai Dixian pills high-, medium-dose group and Shexiang Baoxin pills group also significantly increased (P< 0.01, P<0.001). Compared with sham operation group, VEGF and bFGF mRNA expression of model group, Tongmaidixian pills high-, medium-dose group and Shexiang Baoxin pills was increased (P<0.05, P< 0.01). Compared with model group, VEGF and bFGF mRNA expression of Tongmaidixian Pills high-, medium-dose group and Shexiang Baoxin pills group significantly increased (P<0.01, P< 0.001). Conclusion The effect of Tongmai Dixian pills is closely related to vascular endothelial growth factor (VEGF, bFGF), and play an important role in the ischemic myocardial angiogenesis.%目的 观察通脉地仙丸对大鼠缺血心肌血管生成及其相关基因表达的影响.方法 除假手术组外,大鼠进行慢性心肌缺血造模,造模成功后,随机分为模型组、麝香保心丸组与通脉地仙高、中、低剂量组,各组灌胃给药,6周后采用免疫组化法检测心肌组织中血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)蛋白含量,并观察VEGF、bFGF mRNA的表达.结果 与假手术组比较,模型组、通脉地仙丸各剂量组和麝香保心丸组bFGF、VEGF蛋白表达上调(P< 0.05,P<0.01,P<0.001);与模型组比较,通脉地仙各剂量组和麝香保心丸组bFGF、VEGF蛋白表达均上调(P<0.05,P<0.01,P<0.001);与假手术组比较,模型组、麝香保心丸组和通脉地仙高、中剂量组VEGF、bFGF mRNA表达上调(P<0.05,P<0.01);与模型组比较,通脉地仙高、中剂量组和麝香保心丸组VEGF、bFGF mRNA表达均上调(P<0.01,P<0.001).结论 通脉地仙丸对大鼠缺血心肌血管生成的影响与血管内皮生长因子VEGF、bFGF相关,且发挥重要作用.
