目的 通过肥大细胞(MC)与五羟色胺(5-HT)在肠易激综合征(IBS)大鼠肠道组织的表达改变,探讨九香止泻片的作用机制.方法 选用40只SD大鼠,随机分为空白组、模型组、实验组、对照组,每组10只.除空白组外,其余各组均予以胃湿热方法造模,同时再予以IBS慢性应激造模.造模至第22日开始,实验组予以九香止泻片灌胃,原药材量12.27 g/(kg·d);对照组予盐酸洛哌丁胺灌胃1.08 mg/(kg·d),每日1次,连续灌胃7 d.实验结束时观察各组大鼠肠道中MC与5-HT的表达等改变.结果 实验组在抑制湿热泄泻型IBS大鼠肠道中MC与5-HT表达等方面与模型组比较,差异有统计学意义(P<0.05).结论 九香止泻片能够抑制湿热泄泻型IBS模型大鼠肠道(回盲部,结肠远端)中的5-HT及Mc的异常表达,从而达到止泻的目的.%Objective To explore the mechanism of Nine Enteric tablets by observing the mast cells and 5-HT expression of irritable bowel syndrome (IBS) rats. Methods Forty SD rats were divided into four groups: blank group, model group, experimental group, control group, 10 in each group. Except for the blank group, other groups were made for stomach damp-heat, and then for IBS with chronic stress. The experimental group was administrated with Nine Enteric tablets 12.27 g/(kg ·d) and the control group with loperamide hydrochloride 1.08 mg/(kg· d) once per day for 7 d. The intestinal mast cells and 5-HT expression were observed. Result Experimental group inhibited intestinal mast cells and 5-HT expression of damp-heat diarrhea type IBS rats compared with model group, the difference was significant (P <0.05).Conclusion Nine Enteric tablets can inhibit abnormal expression of intestinal (ileocecal, colon distal) mast cells and 5-HT of damp-heat diarrhea type IBS rat model, and thus achieve the purpose of relieving diarrhea.
