异补骨脂素对小鼠胚胎前成骨细胞胶原的影响

         

摘要

目的 探讨不同浓度异补骨脂素干预对小鼠胚胎前成骨细胞(MC3T3-E1)增殖、Ⅰ型骨胶原及AP-1亚基c-Jun表达水平的影响及其与原发性骨质疏松症的关系.方法 细胞分为正常对照组与不同浓度异补骨脂素组.分别采用MTT法检测成骨细胞(MC3T3-E1)增殖、天狼星红染色法测定细胞层的胶原含量、Western-Blot检测c-Jun及RT-PCR检测Ⅰ型胶原mRNA的表达.结果 不同浓度异补骨脂素刺激,可明显促进MC3T3-E1增殖,细胞层胶原含量增加,c-Jun蛋白表达逐渐增强及Ⅰ型胶原mRNA的表达增高,差异均具有统计学意义(P<0.05).结论 异补骨脂素可能通过促进核转录因子c-Jun表达,进而促进骨胶原合成,从而发挥其预防与治疗骨质疏松症的作用.%Objective To evaluate the impact of different concentrations of isopsoralcn on mouse preosteoblasts (MC3T3-E1) proliferation, type Ⅰ collagen and c-Jun subunit of AP-1 transcription factor,and to describe the relation with primary osteoporosis.Methods Cells were divided into normal control group and different concentrations of isopsoralen groups.Cells proliferation were detected by MTT.The content of cells layer collagen were detected by sirius red method.Western-Blot was used to detect the c-Jun subunit of AP-1.RT-PCR was used to detect mRNA expression of type Ⅰ collagen.Results Different concentrations of isopsoralen stimulated groups promoted the proliferation of MC3T3-E1 cells, cells layer collagen content were increased, the expression of c-Jun protein and type Ⅰ collagen mRNA were increased, and all the differences were statistically significant (P<0.05).Conclusion Isopsoralen may has the mechanism of promoting the expression of nuclear transcription factor C-jun and then stimulating bone collagen synthesis, thus have the function of prevention and treatment of osteoporosis.
