白念珠菌 FLO8基因突变株构建及鉴定

摘要

Objective To construct Candida albicans FLO8 mutations.Methods We inserted Candida albicans FLO8 gene under the control of the ADH1 promoter in pCP20 plasmid,and plasmids containing the point mutations FLO8 R209T , FLO8 A3 1 1 T ,FLO8 6 54Ter ,FLO8 G723R and FLO8 T7 5 1 D were constructed using site-directed mutagenesis from plasmid pCP20.Then we introduced the inserts with FLO8 point mutations into Candida albicans SN1 52 flo8/flo8 at the endogenous ADE2 gene loci by homologous recombination.Results The plasmids with FLO8 point mutations were constructed successfully by DNA sequencing;The inserts with FLO8 point mutations were exactly recombined to the ADE2 gene loci of Candida albicans SN1 52 flo8/flo8 as confirmed by PCR.Conclusions Candida albicans FLO8 mutations can be efficiently constructed with pCP20 plasmid by site-directed mutagenesis,homologous recombination,etc.%目的：构建白念珠菌 FLO8基因突变株。方法将白念珠菌 FLO8基因插入 pCP20质粒载体 ADH1启动子之后，通过定向诱变获得 FLO8基因 R209T、A311T、654Ter、G723R、T751D 突变质粒载体，再通过同源重组技术将带有FLO8突变的基因片段整合至 SN152flo8/flo8菌株的 ADE2位点。结果通过测序鉴定 FLO8基因突变质粒载体构建成功；通过 PCR 验证表明突变 FLO8基因整合到 SN152flo8/flo8菌株的 ADE2位点。结论以 pCP20质粒为载体，通过定向诱变、同源重组等技术，可以高效构建白念珠菌 FLO8基因突变株。
