Ndrg2蛋白在匹罗卡品诱导癫痫大鼠模型的表达规律

摘要

Objective To test a hypothesis that the protein expression of N-Myc downstream regulated gene 2 (Ndrg2) may be enhanced in the hippocampus of rat models of pilocarpine-induced epilepsy and explore the pattern of Ndrg2 expression. Methods Twenty-eight rats were recruited and randomly divided into 2 groups: 24 rats were enrolled in experimental group while the others in controls.The rat models in the experimental group were induced by lithium-pilocarpine protocol. Then survival rats were sacrificed 4 and 24 h, and 7 d and 6 weeks after the onset of status epilepticus (SE) while the controls were sacrificed 24 h after vehicle treatment. Temporal and spatial protein expression characteristics of Ndrg2 in the hippocampus were analyzed by immunofiuorescence labeling. Results The protein level of Ndrg2 increased by 62% and 37% on the 7th d and in the 6th weeks of onset of SE,respectively; statistically significant differences were noted between the experimental group at the above time points and the control group (P<0.05). Ndrg2 immunoreactive cells were concentrated along the subgranular zone of the hippocampus. Conclusion The protein expression of Ndrg2 increases significantly in the hippocampus of rat models with chronic epilepsy, with its location mainly in perinuclear region ofastrocyte, which might result from astrogliosis activated by SE.%目的 验证Ndrg2蛋白在匹罗卡品诱导癫痫大鼠模型的海马区表达上调这一假说,并探索其表达规律.方法 28只SD大鼠采用完全随机数字表法分为2组:实验组24只,对照组4只.实验组大鼠建立锂-匹罗卡品癫痫大鼠模型,将成功建模并存活的致痫鼠按癫痫持续状态(SE)后4 h、24 h、7 d和6周共4个时间点杀死取脑,对照组大鼠给予生理盐水处理后同时间点取样.运用免疫荧光双标的方法分析海马区Ndrg2蛋白的时间空间表达规律.结果 Ndrg2蛋白在SE后7 d表达增长了62%,在SE后6周增长了37%,与对照组比较差异均有统计学意义(P<0.05).Ndrg2免疫反应阳性细胞集中排列于海马颗粒层下带.结论 Ndrg2蛋白表达在慢性癫痫大鼠模型中显著增加,集中表达于星形胶质细胞核周区.其原因推测为SE启动的星形胶质细胞增生硬化.
