PCR法检测弓形虫感染家兔血液中弓形虫基因片段

摘要

目的 用PCR法检测弓形虫感染家兔血液中弓形虫基因片段.方法 弓形虫RH株速殖子感染雄性家兔16只,在感染前后耳缘静脉采集血液标本,-40℃保存.用普通PCR检测血液标本中虫体DNA基因片段.结果 普通PCR检测感染家兔总阳性率为45%.感染后第1周检出率仅22%,高峰期维持在感染后2~4周,检出率约为60%左右.其后检出率下降为25%左右并一直维持.结论 采用血液作为样品进行弓形虫PCR检测时其最佳检测时间为感染后1~4周,其后检出率逐渐下降.%Objective To detect toxoplasma gondii(T.g)DNA fragments in blood from infected rabbits by multiplex polymerase chain reaction(PCR).Methods Sixteen rabbitts were infected with T.g tachyzoites by intraperitoneal injection.The samples of blood were respectively collected from the rabbits'helix vein vessels and storded at -40℃.Toxoplasma gondii DNA was detected by PCR.Results The positive rate of all blood samples fromthe infected rabbits was 45% and that of the blood samplesin 1 week after infection only was 22%.However.it was showed that the positive rateof the blood samples was about 60% in 2~4 week after infection and that was the highest in all of the infection period.Then the positive rate descended and was maintained about 22% after 4 week.Conclusions The optimal time of detection of T.g DNA in blood by PCR is in 1~4 week after infection and the positive rate is lower after that period.
