反义寡脱氧核苷酸逆转人肝癌多药耐药性的实验研究

摘要

Objective To study the role of antisense phosphorothioateoligodeoxynucleotides (ASPODNs)in reversal of multidrug resistance(MDR) of human lepatic cancer cell line. Methods The reverse rate of ASPODNs to MDR was analysed by MTT assay and the effect of ASPODNs on MDR gene expression at mRNA by RT-PCR and at proteins by flow cytometry. Results ASPODNs almost completely inhibited related MDR gene′s expression.This inhibition was time-effective and dose-effective. The reverse rates of ASPODNs to MDR induced by mdr1,MRP,LRP and CSTp1 were 90.20%-100%. The reverse rate of ASPODNs to complex MDR was 70.09%-100%. Conclusion Clinical application of a combination of several ASPODNs to revert MDR is feasible.%目的 探讨反义寡脱氧核苷酸 (ASPODNS)对人肝癌多药耐药基因(MDR)的逆转作用。 方法ASPONs作用于肝癌MDR细胞模型后,用MTT法检测其逆转作用,用逆转录PCR和流式细胞术检测其对4种MDR基因表达的影响。 结果 ASPODNS抑制MDR基因的表达存在时间-效应和剂量-效应关系,对单基因MDR细胞耐药性的逆转率为90.26%~100%,对多重MDR细胞耐药性逆转率为70.09%~100%。 结论 多种ASPODNs联合逆转多重MDR具有可行性并取得明显效果。
