Tyro3受体酪氨酸激酶调控糖蛋白Ⅵ介导的血小板活化

         

摘要

Objective To study the role of Tyro 3 tyrosine kinases receptor in glycoprotein VI (GPVI)-mediated platelet activation. Methods Platelets were isolated from wild type ( WT) and Tyro 3 knockout (Tyro3-/- ) mice,then we examined their responses to GPVI stimulation by measuring integrin allb|33 activation .granule release,aggregation and spreading. Results Compared with WT platelets,Tyro 3-deficient platelets displayed a suppressed aggregation and spreading after CPVI stimulation, a reduced CD 62P expression and integrin allb|33 activition also be observed. Conclusion Tyro 3 plays an important role in CPVI,mediated platelet activation.%目的 研究Tyro3受体酪氨酸激酶在糖蛋白Ⅵ(glycoprotein Ⅵ,GPVI)介导的血小板活化中的调控作用.方法 以Tyro 3基因敲除小鼠的血小板为研究对象,应用GPVI的特异性刺激剂,在体外检测血小板的聚集和铺展能力,并应用流式细胞术分析血小板的颗粒释放及整合素αIIbβ3的活化情况.结果与野生型小鼠相比,Tyro 3基因敲除小鼠血小板的聚集和铺展能力减弱,颗粒释放标记物CD 62P的数量减少,整合素αⅡbβ3的活化也受到明显抑制.结论Tyro 3受体酪氨酸激酶在糖蛋白Ⅵ介导的血小板活化中具有重要的调控作用.
