靶向心肌声学造影监测兔心肌缺血或再灌注损伤中IL-8的表达

         

摘要

Objective To investigate the effect of targeted myocardial contrast echocardiography (MCE) in monitoring the expression of IL-8 in myocardial ischemia/reperfusion injury (MIRI) of rabbit.Methods The MCE examination was employed by injecting SonoVue and IL 8 monoclonal antibody microbubble.The expression of IL-8 was detected by using QPCR.Results The intensity of myocardial ischemia (VI) of the group of targeted MCE was increased with the prolongation of reperfusion time and reached the peak at 180 min after reperfusion.The values of VI in myocardium ischemic area of targeted MCE group were significantly higher than those of normal MCE group at 30-180 min after reperfusion.Correlation analysis showed that the target MCE results were positively correlated with QPCR (P<0.05).Conclusions Targeted MCE can non-invasively monitor the expression of IL-8 in MIRI,increase the sensitivity of MIRI diagnosis,and evaluate myocardial reperfusion.%目的 探讨靶向心肌声学造影(MCE)监测兔心肌缺血或再灌注损伤(MIRI)中IL-8表达的效果.方法 经兔耳缘静脉注射SonoVue及携IL-8单抗超声微泡行MCE检查,实时荧光定量PCR (QPCR)检测IL-8的表达量.结果 携IL-8单抗微泡组(靶向MCE组)随着再灌注时间延长心肌缺血区声强度(VI)逐渐增高,于再灌注180 min达高峰;再灌注30~180 min各时段靶向MCE组心肌缺血区VI值明显高于SonoVue微泡组(普通MCE组).相关性分析得出靶向MCE结果与QPCR结果呈正相关(P<0.05).结论 靶向MCE可无创性地监测MIRI中IL-8的表达量,提高对MIRI诊断的灵敏度,以及评价心肌再灌注情况.
