超声辐照携重组人血管内皮抑制素脂质微泡抑制鸡胚尿囊膜血管生成研究

         

摘要

Objective To investigate the antiangiogenic effects in chick chorioallantoic membrane (CAM) of recombinant human endostatin loaded lipid microbubbles destructed by ultrasound.Methods The recombinant human endostatin was linked to phospholipid using Biotin-Avidin system to prepare the drug-loaded microbubbles,the diameter and drug-loading rate were measured,and the in vivo antiangiogenic analysis was performed.Results The prepared microbubbles were uniform in size with a mean diameter of (3.0±0.4)μm and drug-loading rate of 0.8 mg/108microbubbles,emitted green fluorescence under fluorescence microscope.In the group of ultrasound destruction of drug-loaded microbubbles,both new blood vessels area and vascular area/total area ratio of CAM were the least among all the groups,and the differences were significant.Conclusions Ultrasound-targeted destruction of recombinant human endostatin loaded microbubbles can significantly inhibit angiogenesis of chick chorioallantoic membrane.%目的 探讨超声波辐照击破携重组人血管内皮抑制素脂质微泡对鸡胚尿囊膜血管生成的影响.方法 采用生物素-亲和素系统制备携重组人血管内皮抑制素脂质微泡,测定微泡粒径、载药量;采用鸡胚尿囊膜模型观察超声辐照载药微泡对血管生成的抑制作用.结果 制备的载药微泡平均粒径(3.0±0.4) μm,荧光显微镜可见FITC标记的微泡表面发出绿色荧光,ELISA测得每108微泡载药量为0.8 mg;超声辐照载药微泡治疗组鸡胚尿囊膜新生血管面积、血管面积/总面积比值均小于其余各组(P<0.05).结论 超声波辐照击破携重组人血管内皮抑制素脂质微泡具有明显抑制鸡胚尿囊膜血管生成作用.
