子痫前期胎盘组织 TGF-β1高表达对滋养细胞凋亡的影响

         

摘要

目的:探讨转化生长因子β1( TGF-β1)在正常妊娠和子痫前期患者胎盘组织中的分布和表达及与滋养细胞凋亡的关系。方法采用免疫组织化学法检测30例子痫前期(轻、重度)和10例正常孕妇胎盘中TGF-β1的分布和表达,采用TUNEL法检测3组胎盘滋养细胞凋亡的情况。结果 TGF-β1在正常妊娠和子痫前期患者胎盘的滋养细胞、内皮细胞、基质细胞中均有表达,以滋养细胞表达为主。 TGF-β1在对照组、实验组1和实验组2的积分光密度逐渐增高,实验组1与对照组比较差异有统计学意义(t=10.834,P<0.01);实验组2与实验组1比较差异有统计学意义(t=13.056,P<0.01)。正常妊娠和子痫前期患者胎盘的滋养细胞、内皮细胞、基质细胞均存在凋亡,以滋养细胞凋亡为主。实验组1和组2均较对照组凋亡率明显增高(χ2值分别为18.42、48.87,均P<0.01);随着病情加重,实验组1、实验组2细胞凋亡率呈上升趋势(χ2=7.70,P<0.01)。滋养细胞TGF-β1表达与胎盘绒毛细胞凋亡率在实验组2、实验组1均具有相关性(r值分别为0.910、0.520,均P<0.01)。结论子痫前期患者胎盘组织中的TGF-β1高表达可能与其滋养细胞凋亡增加有关。%Objective To research the expression and distribution of transforming growth factor-beta ( TGF-β1 ) in placental tissue of normal pregnant women and preeclampsia patients and their correlation with the apoptosis of trophocyte .Methods The expression and distribution of TGF-β1 in placental tissue of 30 preeclampsia patients ( mild and severe cases ) and 10 normal pregnant women were detected with immunohistochemistry technique ( IHCT) .The apoptosis of placenta trophocyte cells was tested using TUNEL method .Results TGF-β1 was detected expressing in trophocyte , endothelial and stroma cells in normal pregnant women and preeclampsia patients , primarily in trophocyte cells .The integrated optical density of TGF-β1 increased gradually in the control group , experimental group 1 and experimental group 2.The difference between experimental group 1 and the control group was statistically significant (t=10.834, P<0.01), and there was significant difference between experimental group 2 and experimental group 1 (t =13.056,P <0.01).The apoptosis existed in trophocyte , endothelial and stroma cells , primarily in trophocyte cells , in normal pregnant women and preeclampsia patients .The apoptosis rate of experimental group 1 and experimental group 2 was higher than that of the control group (χ2 value was 18.42 and 48.87, respectively, both P<0.01).With the aggravating of disease, there was a rise trend in apoptosis rate of experimental group 1 and experimental group 2 (χ2 =7.70,P<0.01) .The expression of TGF-β1 in trophocyte was related with the apoptosis in placental villus cells in experimental group 1 and experimental group 2 (r value was 0.910 and 0.520, respectively, both P<0.01).Conclusion The high expression of TGF-β1 in the placental tissue of preeclampsia patients might be related with the increase of trophocyte apoptosis .
