生物大分子高分辨率冷冻电镜三维重构技术

         

摘要

The 3D reconstruction technology of the cryo-electron microscopy (cryo-EM)can be used to analyze the high-resolution 3D structure of the biological macromolecular complex. The cryo-EM does not require the sample crystallization,the applicable biological sample is more extensive,and it can maintain the biological sample near the physiological state.In recent years,with the development of the electron microscopy hardware and computer image processing software technology,the structural analytical resolution by the cryo-EM has reached the atomic level and has gradually become one of the mainstream methods for the biological structure analysis.This paper introduces the origin, development and future trend of the 3D reconstruction technology of the cryo-EM.%冷冻电子显微三维重构技术(简称冷冻电镜)能够解析生物大分子复合体的高分辨率三维结构.冷冻电镜不需要样品结晶,适用的生物样品较为宽泛,且保持了生物样品的近生理状态.近年来随着电镜硬件和计算机图像处理软件技术的发展,冷冻电镜结构解析分辨率已经达到原子水平,逐渐成为生物结构解析的主流方法之一.该文介绍了冷冻电镜三维重构技术的起源、发展及未来趋势.
