响应面法优化木瓜蛋白酶提取工艺

         

摘要

Using fresh papaya as raw material. Extraction of papain on ultrafiltration choicing the L-cys, ED-TA, pressure, temperature, time, solid-liquid ratio, six factors as single factor experiment, the extraction of papain by ultrafiltration to determine the optimum process conditions, the L-cys and EDTA for the enzyme pro-tective agent optimal concentrations were:0.04 mol/L and 0.002 mol/L. The selected temperature, pressure, time and ratio of material to liquid four conditions by response surface experiments, mathematical model, and regression analysis, evaluation model, the results showed that the optimum conditions of ultrafiltration extrac-tion of Papain is:temperature 55℃, pressure 0.25 MPa, time 3.7 min, material density 30%, the enzyme ac-tivity is (9.31±0.27 )(U×10 000)/mg, it is close with the model validation activity of 9.355 (U×10 000)/mg. That response surface method can be used to optimize the extraction process of papain better.%采用新鲜的番木瓜为原料,探究超滤法提取木瓜蛋白酶的最佳工艺条件,选定L-cys、EDTA、温度、压力、时间、物料浓度6个因素进行单因素试验,确定超滤法提取木瓜蛋白酶的较佳工艺条件,其中L-cys和EDTA为木瓜蛋白酶的保护剂,其浓度分别在0.04 mol/L和0.002 mol/L时对木瓜蛋白酶的酶活性保护最强。选定超滤过程中的温度、压力、时间、物料浓度4个条件进行响应面试验,建立数学模型,然后回归分析,模型评价,结果表明超滤法提取木瓜蛋白酶的最佳条件为:温度55℃,压力0.25 MPa,时间3.7 min,物料浓度30%,此时酶活性大小为(9.31±0.27)万单位/mg,与模型验证的酶活性9.355万单位/mg非常接近。说明响应面法能较好的用于木瓜蛋白酶提取工艺的优化。
