普安银鲫早期发育中 ACCα和 FAS 基因表达及外源物质的影响

         

摘要

In order to discuss the gene expression characteristic of acetyl-CoA carboxylaseα( ACCα) and fatty acid syn-thase ( FAS) , and effects of glucose solution, vitamin C solution on them during early development of Carassius auratus gibelio were studied.According to the results of preliminary experiment, membrane breaktime and hatching rate were recorded to decide the optimal concentrations of glucose and vitamin C.Solutions without addition ( control group) and with optimal concentrations of glucose (15 g/L glucose group), concentrations of vitamin C (30 mg/L vitamin C group) were used for hatching, and the gene expression characteristics of ACCαand FAS were analysed.The real-time PCR technology was used to examine the mRNA expression level of ACCαand FAS genes and their mRNA expression levels in the yolk-sac larva of C.auratus gibelio, and the effects of glucose and vitamin C on the mRNA expressions of these two genes were explored.The results showed that ACCαand FAS genes were widely expressed in endogenous nutrition stage, mixed nutrition stage and exogenous nutrition stage of C.auratus gibelio.The mRNA expression level of ACCαand FAS showed“down-up” trend.In the group of glucose (15 g/L), the mRNA expression level of ACCαand FAS increased, and its significantly higher than that of control group during yolk-sac larva development of C.auratus gibelio ( P<0.05 ).In the group of vitamin C ( 30 mg/L) , the mRNA expression level of ACCαand FAS genes increased, and its significantly higher than that of control group in mixed nutrition stage and exogenous nutrition stage ( P<0.05 ).The research showed that the lipid metabolism synthesis decreased in mixed nutrition stage of C.auratus gibelio.Appropriate glucose solution (15 g/L) could induce the mRNA expression of ACCαand FAS genes.%为了探讨普安银鲫( Carassius auratus gibelio)早期发育中乙酰辅酶A羧化酶α( acetyl-CoA carboxylas eα, ACCα)和脂肪酸合成酶( fatty acid synthase, FAS)表达特点及葡萄糖和维生素C溶液分别浸泡对它们的影响,采用荧光定量PCR技术检测ACCα和FAS基因在普安银鲫卵黄囊期的表达情况及mRNA表达水平,并检测了葡萄糖、维生素C对这两种基因mRNA表达量的影响。结果显示, FAS和ACCα基因在普安银鲫内源营养期、混合营养期和外源营养期均有表达。普安银鲫卵黄囊期ACCα和FAS mRNA表达量呈“下降-上升”变化。在15 g/L葡萄糖处理组中,从内源营养期发育到外源营养期, FAS和ACCα的mRNA表达量呈上升变化,且2种基因的表达均显著高于对照组(P<0.05)。在30 mg/L维生素C处理组中, FAS和ACCα的mRNA表达量呈上升变化,且在混合营养期和外源营养期,2种基因的表达均显著高于对照组(P<0.05)。研究表明,普安银鲫发育至混合营养期时,机体内脂质合成代谢减弱。适宜水平的葡萄糖和维生素C均能诱导ACCα和FAS mRNA的表达,有利于鱼体内脂质的合成。
