摘要

Objective To observe the effect of mengoferin monosodium salt on insulin resistance in mice with type 2 diabetes mellitus.Methods Intraperitoneal injection of streptozocin (STZ, 150 mg/kg) combined with high fat diet feeding was given to mice to induce a pathological model of insulin resistance type 2 diabetes .The influence of mengo-ferin monosodium salt (50 and 100 mg/kg were given by gavage once a day for 2 weeks) on the fasting plasma glucose (FBG), fasting insulin (FINS), oral glucose tolerance (OGTT), serum low density lipoprotein (LDL-C) and triglycer-ide (TG) levels, as well as the mRNA expression of glucose transporter -4 (GLUT4) and insulin receptor (InsR) were evaluated.Results Compared with the model group , the serum level of FBG , TG, LDL-C were significantly reduced by low and high-dose of mengoferin monosodium salt in insulin resistance mice .Besides, mengoferin monosodium salt in-creased insulin sensitivity , improved sugar tolerance and ISI , and lowered HOMA-IR index.The expression of GLUT4 and InsR mRNA were also up-regulated by the treatment of mengoferin monosodium salt .Conclusion Mengoferin mon-osodium salt can improve glycolipid metabolism , increase insulin sensitivity in type 2 diabetes and insulin resistance .%目的:研究芒果苷单钠盐对链脲佐菌素( STZ)诱导2型糖尿病小鼠及其胰岛素抵抗的影响。方法昆明小鼠60只,随机分为5组:正常组,模型组,二甲双胍组,芒果苷单钠盐低剂量组、高剂量组,每组12只。除正常组外,其余各组均采用高脂饲料联合腹腔注射STZ (150 mg/kg )建立胰岛素抵抗2型糖尿病小鼠模型,成模后,每天给予芒果苷单钠盐(50、100 mg/kg)进行治疗,连续14 d,观察小鼠体重、空腹血糖( FBG)的变化情况。末次给药后禁食12 h,测定口服糖耐量( OGTT)。眼球取血,检测血清胰岛素、三酰甘油( TG)和低密度脂蛋白胆固醇(LDL-C)的含量,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR);RT-PCR法测定骨骼肌葡萄糖转运蛋白4(GLUT4)mRNA和肝脏胰岛素受体(InsR)mRNA的表达水平。结果与模型组相比,芒果苷单钠盐高、低剂量组小鼠的FBG、胰岛素和HOMA-IR均明显降低,OGTT、ISI显著升高;GLUT4和InsR mRNA的表达水平出现显著的上调;TG、LDL-C含量也有一定程度的降低。结论芒果苷单钠盐通过改善糖脂代谢、提高胰岛素敏感性,发挥其治疗糖尿病及胰岛素抵抗的作用。
