人参皂苷Rb1对心力衰竭大鼠心肌细胞葡萄糖转运载体-4的影响

         

摘要

Objective On the basis of adriamycin - induced chronic heart failure ( CHF) , to elucidate the effect of ginsenosides - Rbl ( Gs - Rbl ) on improving cardiac function via affecting glucose transporter -4 ( GLUT -4) in vivo and in vitro. Methods CHF rats were induced with adriamycin and randomly divided into control group (n = 10) , adriamycin group (n = 15) and Gs - Rbl group [70 mg/(kg·d) , n = 17]. The neonatal rat cardiomyocytes were randomly divided into control group, adriamycin group (1 μmol/L) and Gs - Rbl group (200 μmol/L) . At the 4th week, the left ventricular ejection fraction (LVEF) was analyzed by echoeardiography, as the expression of GLUT -4 were assessed by Western blot and RT - PCR. Results Compared to adriamycin group, Gs - Rbl significantly improved LVEF ( P = 0. 015) . Both protein and mRNA of GLUT -4 were significantly up - regulated in Gs - Rbl group than those in adriamycin group, but significantly lower than those in control group in vivo; so was in vitro. The expression of GLUT - 4 protein in the membrane of cardiomyocytes was significantly lower in adriamycin group than those in Gs - Rbl group and control group. Conclusion The inhibition of both expression and transformation of GLUT -4 involve in the pathogenesis of adriamycin - induced CHF, which could be partially reversed by Gs - Rbl.%目的 探讨人参皂苷Rb1(ginsenoside Rb1,Gs-Rb1)减轻阿霉素所致的慢性心力衰竭(CHF)效应与改善葡萄糖转运载体-4(glucose transporter-4,GLUT-4)的关系.方法 阿霉素诱导CHF大鼠被随机分为对照组(n=10)、阿霉素组(n=15)和Gs-Rb1组[70 mg/(kg·d),n=17],同时培养的乳鼠心肌细胞随机分为对照组、阿霉素组(1 μmol/L)和Gs-Rb1组(200 μmol/L).干预完毕后,检测心脏超声、GLUT-4蛋白和mRNA水平.结果 (1)与阿霉素组相比,Gs-Rb1组的左室射血分数显著提高(P<0.05);(2)在体内研究中,Gs-Rb1组GLUT-4蛋白和mRNA水平均显著高于阿霉素组(P<0.05),但显著低于对照组(P<0.05);(3)在体外研究中,与阿霉素组相比,Gs-Rb1组GLUT-4 蛋白水平和mRNA水平显著提高(P<0.05),但均显著低于对照组(P<0.05);(4)阿霉素组心肌细胞膜GLUT-4蛋白水平显著低于对照组(P<0.05),而Gs-Rb1组心肌细胞膜GLUT-4蛋白显著高于阿霉素组(P<0.05).结论 在阿霉素促发CHF进程中,GLUT-4的表达和移位被抑制可能起着一定作用;Gs-Rb1改善阿霉素CHF效应与促进GLUT-4的表达和移位有关.
