非诺贝特对高糖诱导的人视网膜微血管内皮细胞增殖和细胞周期的影响

         

摘要

目的 探讨非诺贝特对高糖诱导的人视网膜血管内皮细胞的增殖及细胞周期的影响.方法 将人视网膜血管内皮细胞分为正常组(5.5 mmol/L葡萄糖)、高糖组(25 mmol/L葡萄糖)、高糖+不同浓度(10、50、100 μmol/L)的非诺贝特组.采用CCK-8法检测人视网膜微血管内皮细胞增殖率,采用Hoechst33258染色观察细胞凋亡形态学改变,采用流式细胞术检测细胞周期改变.结果 CCK8结果显示:与正常组相比,高糖组人视网膜微血管内皮细胞增殖能力增加,差异有统计学意义(P<0.001);与高糖组相比,各浓度非诺贝特组人视网膜微血管内皮细胞增殖能力下降,差异也均有统计学意义(均P<0.000);Hoechst33258染色结果显示:染色后荧光显微镜下观察:非诺贝特组细胞出现核固缩、核碎裂;流式细胞仪检测结果显示:非诺贝特组细胞发生G1/S期阻滞(均P<0.05).结论 非诺贝特能够抑制人视网膜微血管内皮细胞增殖,其机制可能是通过影响细胞周期调节并促进细胞凋亡.%Objective To investigate the effects of fenofibrate on cell proliferation and cell cycle of human retinal capillary endothelial cells ( HRECs) in high glucose environment. Methods The HRECs were divided into 5 groups: control group ( Group N) , high glucose group ( Group H) , and treatment groups (high glucose with different concentra -tions of fenofibrate). CCK8 assay, Hoechst33258 staining and flow cytometry were used for assessment of cell prolifera -tion, chromatin distribution and cell cycle , respectively. Results To proliferation of HRECs in Group H was significantly higher than that in Group N (P < 0. 001). However, this effect was significantly inhibited by fenofibrate treatment (P < 0. 000). Typical apoptosis and significant G1/S blocking were observed (P <0. 05). Conclusion Fenofibrate suppresses the proliferation of HRECs in vitro by affecting the cell cycle and promoting cell apoptosis .
