目的 研究三甲基氯化锡(trimethyltin chloride,TMT)对大鼠脉络丛上皮细胞钠-钾-ATP酶(NKA)活性的影响.方法 取SPF级SD雌性大鼠,TMT 10mg/kg腹腔注射染毒,染毒后1、24、36、48、72 h处死,分别测定血钾、脉络丛NKA活性、脑脊液(CSF)电解质和葡萄糖(GLU)浓度,并在光镜下观察脉络丛上皮细胞形态.结果 染毒动物出现低血钾,并伴有神经损害症状.染毒动物脉络丛NKA的活性显著下降,与血钾浓度变化一致.染毒24、36 h动物CSF中GLU升高,而染毒48、72 h动物CSF中钾显著下降.光镜下未发现脉络丛上皮细胞形态异常.结论 TMT可抑制脉络丛上皮NKA的活性,引起CSF电解质失衡,这可能是TMT引起中枢神经系统损伤的机制.%Objective To study the inhibitive effects of trimethyltin chloride ( TMT ) on the activity of Na+-K+ - ATPase ( NKA ) on choroid plexus epithelial cells in SD female rats. Methods The rats in treatment groups were in-traperitoneally injected with TMT ( 10 mg/kg ) and sacrificed after 1 h, 24 h, 36 h, 48 h and 72 h, respectively. The NKA activity on the choroid plexus epithelial cells and serum K+ were assessed. Cerebrospinal fluids ( CSF ) were collected for assessment of Na+ , Cl- ions and glucose concentrations, while the morphological observation of the choroids plexus was carried out in optical microscope. Results Sustained hypokalemia and neurological damage symptoms were presented after TMT poisoning. The decrease of serum K+ was consistent with the reduction of NKA activitiy on choroid plexus. Significant elevation of glucose was observed at 24 h and 36 h, with significant descence of CSF K+ revealed at 48 h. No morphological change was observed in choroid plexus epithelial cells under light microscope. Conclusion The TMT inhibits the activity of NKA on choroid plexus, so as to induce CSF electrolyte imbalance, suggesting a posible mechanism of TMT-induced central nerve system injury.
