Objective To study the frequencies of allele and genotype distribution of Neurite growth inhibitor - A ( Nogo - A) gene intron single nucleotide polymorphism ( SNP) in Guangxi population, and to further analyze the distributions of Nogo - A polymorphism among different races. Methods The Nogo - A gene intron rsl012603 C/T polymorphism was examined by the polymerase chain reaction - single base extension ( PCR - SBE) and DNA sequencing in 199 Chinese people in Guangxi, and compared with other the four populations ( HapMap - CEU, HapMap - YRI, HapMap -JPT and HapMap - HCB) from Human Genome Project group (Hapmap) data. Results Nogo -A genotype frequencies of CC, CT and TT were 71. 4% , 23. 1% and 5. 5 % , respectively, with allele frequencies of C and T were 82. 9% and 17. 1% , respectively, in Chinese Guangxi population. The frequency of genotype of Nogo -A gene rsl012603 C/T polymorphism was significant different between males and females ( P < 0. 05 ) . Meanwhile, the frequencies of allele and genotype distribution of Nogo -A gene rsl012603 C/T polymorphism were significant different with those of HapMap - CEU, HapMap - YRI and HapMap - JPT ( P < 0. 05 ) , though no significant difference was revealed between Guangxi population and HapMap - HCB population (P >0. 05) . Conclusion There is Nogo - A gene polymorphism in Guangxi population, with significant difference to the others ethnic populations.%目的 了解Nogo-A基因内含子区单核苷酸多态性(SNP)各等位基因及基因型在中国广西地区人群中的分布频率,比较其在不同种族间分布的差异.方法 采用单碱基延伸的PCR技术和DNA测序法检测199例中国广西人的Nogo-A基因内含子区rs1012603 C/T多态性,并结合人类基因组计划(Hapmap)公布的4个人群(欧洲人、非洲人、日本人和中国北京人)的SNP 分型数据,比较分析这5个人群的基因型及等位基因的分布频率.结果 Nogo-A基因型CC、CT和TT频率分别为71.4%、23.1%和5.5%;等位基因C、T 频率分别为82.9% 和17.1%.其基因型频率在男女组间比较差异有统计学意义(P<0.05).与欧洲、非洲和日本人群比较,Nogo-A基因多态性的分布频率均差异有统计学意义(P<0.05);而与北京人比较差异无统计学意义(P>0.05).结论 在中国广西人群中存在Nogo-A基因多态性,与其他种族人群比较有差异.
