Objective To explore the methylation abnormality of plasma p73 gene and its clinical significance in patients with colorectal cancer. Methods Totally 142 patients with colorectal cancer were enrolled,including 64 with colorectal cancer(colorectal cancer group)and 78 with colorectal tumor(colorectal tumor group),and 142 healthy volunteers as healthy control group. The methylation of plasma p73 gene was detected by the methylation specific PCR,and the content of serum carcinoembryonic antigen(CEA) was determined by chemiluminescent immunoassay. The methylation of plasma p73 gene and positive rate of serum CEA were compared among the three groups,and the positive rates of plasma p73 gene methylation were compared among the patients with colorectal cancer of different clinical pathological characteristics. Results The positive rates of plasma p73 gene methylation and CEA increased in the order of the control group, the colorectal tumor group and the colorectal cancer group(P<0. 05). Among the patients with colorectal cancer,there were statistically significant differences in the positive rate of plasma p73 gene methylation between different tumor diameters,different infiltrating depths, different organ types,or different clinical stages(all P<0. 05). Conclusion The positive rate of plasma p73 gene is quite high in patients with colorectal cancer. Detection of plasma p73 gene methylation might be valuable in the early diagnosis of colorectal cancer.%目的 探讨结直肠癌患者血浆 p73 基因甲基化异常情况及其临床意义.方法 选择结直肠癌肿瘤患者142例,其中结直肠癌64例(结直肠癌组),结直肠瘤78例(结直肠瘤组),同期选择健康对照组142例.运用甲基化特异性PCR检测血浆p73基因甲基化,应用化学发光法检测血清癌胚抗原(CEA)含量.比较3组血浆p73基因甲基化、血清CEA阳性检出率及不同病理特征的结直肠癌患者血浆p73基因甲基化阳性检出率的差异.结果 3组血浆p73基因甲基化阳性检出率、CEA阳性检出率均呈对照组<结直肠瘤组<直肠癌组(P<0. 05).结直肠癌组患者中, 不同肿瘤直径、浸润深度、组织类型、临床分期者,血浆p73基因甲基化阳性检出率比较,差异均有统计学意义(均P<0. 05).结论 结直肠癌患者血浆p73基因甲基化阳性检出率较高,检测血浆p73基因甲基化可能对早期诊断结直肠癌有较高价值.
