脂联素对高糖培养的大鼠肾小球系膜细胞血管内皮生长因子表达的影响

         

摘要

Objective To observation the effeeL of adiponeetin( ADPM ) on vascular endothelial growth factor ( VEGF ) expressed by high glucose-cultured rat mesangial cell line( IIBZY-1 ),and explore its protective role in diabetic nephropathy mechanism. Methods IIBZY-1 was cultured in vitm, which was randomly divided inLo the following seven groups after the subculture;control group( 5.6 mmol/L glucose ),high glucose group A( 15 mmol/L glucose ),high glucose group B( 30 mmol/L glucose ); Adiponectin group A, B, C, D with the concentration of 2. 5,5,10,20 μg/ml adiponectin ( 30 mmol/L glucose in each group ),respectively ; All the groups were treated for 24 hours. The expressions of VEGF mRNA in cells of each group were measured with RT-PCR,and the protein levels of VEGF were measured in each group wkh ELISA. Results Compared wkh control group, VEGF mRMA and VEGF proLein expression of IIBZY-1 increased in high glucose group A,B( P <0.05 ); VEGF mRMA and VEGF protein expression of IIBZY-1 in each adiponectin group were lower than those in high glucose gmup B( P <(). 05 ). Conclusion Iligh glucose can stimulate the increase of VEGF expression of IIBZY-1 ,and ADPM can inhibit VEGF expression of high glucose-cultured mesangial cells.%目的 观察脂联素对高糖培养的大鼠肾小球系膜细胞表达血管内皮生长因子(VEGF)的影响,探讨脂联素在糖尿病肾病中可能的保护作用.方法 体外培养大鼠肾小球系膜细胞,随机分为7组:对照组、高糖A组、高糖B组(培养液葡萄糖浓度分别为5.6 mmol/L、15 mmol/L、30 mmol/L);脂联素A、B、C、D组(各组培养液葡萄糖浓度均为30 mmol/L+脂联素,脂联素浓度依次为2.5 μg/ml、5 μg/ml、10 μg/ml、20 μg/ml),作用24 h,RT-PCR法测定各组细胞VEGF mRNA表达水平;ELISA法测定各组上清液中VEGF蛋白的含量.结果 与对照组相比,高糖A组、高糖B组大鼠肾小球系膜细胞VEGF mRNA及VEGF蛋白表达升高(P<0.05);脂联素各组大鼠肾小球系膜细胞VEGF mRNA及VEGF蛋白表达低于高糖B组(P<0.05).结论 高糖可刺激大鼠肾系膜细胞VEGF表达增高,脂联素可抑制高糖环境下系膜细胞对VEGF的表达.
