目的 探讨Drosha基因和卵巢癌顺铂耐药的关系.方法 分别采用实时定量PCR和免疫印迹法检测Drosha基因mRNA及其蛋白在A2780/DDP及A2780细胞中表达的差异.结果 A2780/DDP细胞中Drosha基因mRNA及其蛋白的表达量分别是A2780细胞表达量的(56.79±1.65)%和(50.16±1.34)%,两者比较差异均有统计学意义(P<0.001).结论 Drosha基因在A2780/DDP细胞中明显下调,提示Drosha基因和卵巢癌顺铂耐药相关.%Objective To investigate the relationship between Drosha gene and cisplatin( DDP )-resistance in ovarian cancer. Methods The mRNA and protein expression of Drosha gene in A2780/DDP and A2780 were detected by real-time quantitative RT-PCR and Western blot assay, respectively. Results The expression of mRNA and protein of Drosha gene in cisplatin-resistant ovarian cancer cell line A2780/DDP were( 56.79 ± 1. 65 )% and ( 50.16 ± 1. 34 )% , respectively, and the differences were significant compared with those in ovarian cancer cell line A2780( P < 0. 001 ). Conclusion The expression of Drosha gene is down-regulated in A2780/DDP cells,which suggests that Drosha gene is correlated with cisplatin-resistance in ovarian cancer.
