苯巴比妥与牛血清白蛋白的相互作用

         

摘要

为镇静、睡眠类药物的研发提供理论依据,用紫外-可见光谱法和荧光光谱法研究了苯巴比妥(Ph)与牛血清白蛋白(BSA)之间的相互作用。结果表明:Ph 对 BSA 有较强的荧光猝灭作用,其猝灭机理属于静态猝灭,主要靠氢键和范德华力相互作用。Ph 可以被 BSA 运输,结合位置位于 BSA 的亚结构ⅡA中,结合过程中对温度比较敏感,Ph 与 BSA 的结合位点偏向于色氨酸。Mn2+、Cu2+、Co2+对 Ph 与 BSA 结合产生竞争作用,增强药效;Ni2+和 Fe3+对 Ph 与 BSA 结合产生促进作用,延长药效时间。%The interaction between Phenobarbital (Ph) and bovine serum albumin (BSA) was determined by U-V absorption spectrometry and fluorescence spectrometry to provide the theoretical basis for research and development of sedative-hypnotic drugs.The results showed that Ph has the strong fluorescence quenching effect on BSA through interaction between hydrogen bond and VDW force.Ph can be transported by BSA and bound in sub-structure ⅡA of BSA.Ph is sensitive to temperature during the binding process.The binding site of Ph and BSA is partial to tryptophan.Mn2+ ,Cu2+ and Co2+ have the competitive effect during the interaction of Ph and BSA,which can increase medical effectiveness,but Ni2+and Fe3+ have promotion effect on interaction of Ph and BSA,which can prolong time of drug effect.
